The Relationship Between TGF-βRⅡ,Smad4,Smad7 Expressions And Clinicopathological Features In Gastric Carcinoma As Well As Patient Survival

【Objective】To investigate the expressions and significance of TGF-βRⅡ,Smad4,Smad7 protein in gastric carcinoma and determine the relatioship between TGF-βRⅡ,Smad4,Smad7 and tumor clinicopathological features as well as patient survival.In order to evolve the tasks of gastric carcinoma detection and precaution.【Methods】The expressions of TGF-βRⅡ,Smad4,Smad7 in gastric carcinoma, paracancer and normal mucosa,were detected by immunohistochemistry.Analyze the correlation of them,and then analyze relationship between TGF-βRⅡ,Smad4,Smad7 expressions and clinicopathology,as well as patients survival.All statistics analyzed by software of SPSS 15.0.【Results】1 Positive TGF-βRⅡ,Smad4,Smad7 expression was observed in 25.5%,60.7%,47.0%of the gastric carcinoma,respectively.The difference between gastric carcinoma group and control group had statistical difference.2 TGF-βRⅡexpression were found to be statistically associated with tumor differentiation status,the depth of invasion,lymph node metastasisand and Lauren classification(P＜0.05);Smad4 expression were found to be statistically associated with tumor differentiation and Lauren classification(P＜0.05);Smad7 expression were found to be statistically associated with tumor differentiation and Lauren classification (P＜0.05).3 Significant correlation was observed between Smad4 and TGF-βRⅡ(r =0.191, P= 0.007);TGF-βRⅡand Smad7expression(r = -0.160,P = 0.023),respectively. 4 Smad4 positive expression was associated with survival.5 All the factors were analyzed by Cox proportional hazard model.It showed that lymph node metastasisand and the depth of infiltration were independently associated with patients' survival length.【Conclusions】1 TGF-βRⅡwas low expression in gastric carcinoma,the expression was closely related with tumor differentiation status,the depth of invasion,lymph node metastasisand and Lauren classification.2 Smad4 was low expression in gastric carcinoma,the expression was closely related with tumor differentiation status and Lauren classification.3 Smad7 was high expression in gastric carcinoma,the expression was closely related with tumor differentiation status and Lauren classification.4 Significant correlation was observed between Smad4 and TGF-βRⅡ, TGF-βRⅡand Smad7 expression,respectively.5 All the factors were analyzed by Cox proportional hazard model.It showed that lymph node metastasisand and the depth of infiltration were independently associated with patients' survival length.We find that the development of gastric carcinoma is associated with TGF-βRⅡ,Smad4,Smad7.Detect the expression of TGF-βRⅡ,Smad4,Smad7 protein in gastric carcinoma combination will offer important informations about gastric carcinoma.
