ObjectiveLung cancer is one of the most common malignant tumors, the incidence and mortality rates of lung cancer have leapt to the forefront of various types of cancer. In some developed countries like Europe and the United States, the incidence and mortality rates of lung cancer have become the first in a variety of cancers.Tumor metastasis is an important biological character of the malignant tumor.Most cancer patients did not die from primary cancer, but died of metastatic cancer. Therefore, the research of tumor metastasis is an important topic of concern. The occurrence of malignant tumors is the interaction of multiple genes involved in multi-stage process,and tumor metastasis is also a multi-factor role of multiple genes to coordinate the multi-stage process. In recent years, with progress in tumor molecular biology techniques, people have discovered the nm23-H1, TIMP5, KAI1, MRP-1 and other metastasis suppressor genes, and also found a number of tumor metastasis-related genes, such as CD44, c-erb B-2 and so on.ATP-binding cassette transporter E1 (ABCE1) encode a product of 68KD protein called Ribonuclease L (RNase L) inhibitor in multi-cellular cytoplasm of eukaryotic cells,and RNase L can specifically bind and degrade RNA In the cell. ABCE1 protein blocked 2-5A/RNase L pathway through the inhibition of RNase L, thus indirectly inhibiting cell apoptosis. Currently ABCE1 gene and the 2-5A/RNase L pathway of its involved regulation get an expression in the occurrence and progress of certain malignant tumors.Therefore, we have speculated that ABCE1 participate in the progress and metastasis of lung cancer. This experiment will detect the expression differences of ABCE1 gene in primary cancer of lung, metastatic lymph nodes and adjacent tissues through reverse transcription-polymerase chain reaction (RT-PCR) technology and immunohistochemistry (SP), and thus evaluate its impact on progress and metastasis of lung cancer and its clinical significance.MethoedsIn this study we collected 18 cases of specimens about primary cancer of lung, metastatic lymph nodes and adjacent tissues from different resected lung or lung segment. After sending to general pathological examination specimens were divided into two parts, one set immediately frozen in liquid nitrogen,-80℃storage, RT-PCR method for detecting difference of ABCE1 gene expression; another part was fixed immediately by 10% Formalin for immunohistochemistry experiments.ResultRT-PCR results showed that:The expression of ABCE1 gene in lung cancer tissue and metastatic lymph node were higher than adjacent tissues, lung tissues ABCE1 gene expression levels and expression of metastatic lymph nodes was no significant difference (P> 0.05).The immunohistochemistry results showed that:ABCE1 protein expression was mainly located in cytoplasm.Conclusion1.ABCE1 protein was expressed in cytoplasm.2. ABCE1 gene was highly expressed in lung cancer, metastatic lymph nodes.3.ABCE1 may be related to the occurrence and metastasis of lung cancer.
