Esophageal cancer(esophageal carcinoma)is a common digestive system malignant tumor,our country at present is still a high incidence of esophageal cancer,its incidence and mortality is the highest in the world.The main pathological types of esophageal cancer are squamous cell carcinoma and adenocarcinoma.Under the action of a series of factors,the pathological evolution of normal esophageal tissue makes normal cells appear atypia,which is manifested as the excessive proliferation of atypia cells,leading to a series of clinical manifestations of a disease.In recent years,with the continuous improvement of surgical,radiological and chemotherapeutic techniques,the survival of patients with esophageal cancer has been prolonged,but the therapeutic effect is not very significant.With the development of molecular biology,targeted therapy for esophageal cancer has emerged,but its efficacy is not obvious.At present,it is urgent to develop new targeted genes for esophageal cancer,so as to further prolong the survival of patients with esophageal cancer and increase the hope for the cure of patients with esophageal cancer.ABCE1(atp-binding cassette protein E1 or atp-bbillion cassette E1)was first expressed and described by cloning in vitro in 1995 and was named ribonuclease L inhibitor(RLI).As a member of the ABC protein family,ABCE1 has a relatively conservative structure.ABCE1 has become a recognized IFN(interferon)-dependent 2-5a /RNase L antiviral action of the core pathway key enzyme ribonuclease L(RNase L)specific inhibitor,further regulate the stability of cell RNA,and play a universal role.Recent studies have shown that ABCE1 plays a particularly important role in the assembly of chronic primate viruses,especially hiv-1 virus capsids.Further studies have shown that ABCE1 is also involved in the initiation,termination and ribosomal cycle of eukaryotic protein translation.ABCE1 gene is a regulatory gene,which plays a regulatory role in cell proliferation,migration,invasion and apoptosis.It was also found that ABCE1 gene was closely related to the occurrence and development of various malignant tumors.However,no literature has been published on the expression of ABCE1 in esophageal squamous cell carcinoma and its correlation with the clinicopathological features of esophageal carcinoma.This study is the first to explore the relationship between ABCE1 gene and esophageal squamous cell carcinoma,and further explore the relationship between ABCE1 gene and the clinicopathological characteristics of patients with esophageal squamous cell carcinoma,so as to provide a new theoretical basis and research direction for the further study on the occurrence and development of esophageal squamous cell carcinoma.Objective:To investigate the expression of ABCE1 protein in esophageal squamous cell carcinoma and adjacent normal tissues,and to analyze the correlation between ABCE1 protein and clinical features of patients with esophageal squamous cell carcinoma.Methods:1.From December 2017 to April 2019,48 patients with esophageal squamous cell carcinoma(all of whom had not received radiotherapy or chemotherapy before surgery)were selected from the affiliated hospital of chengde medical college.Fresh esophageal squamous cell carcinoma tissues and their corresponding adjacent normal tissues(esophageal tissues more than 5cm away from the tumor margin)were taken as experimental samples.2.Immunohistochemical method was used to detect the expression of ABCE1 protein in 48 cases of esophageal squamous cell carcinoma and adjacent tissues.3.The expression levels of ABCE1 protein in 48 cases of esophageal squamous cell carcinoma and adjacent tissues were detected by Western Blotting(WB).4.Statistical software was used to analyze the correlation between ABCE1 protein and the clinicopathological characteristics(gender,age,alcohol consumption,stage,differentiation and lymph node metastasis)of patients with esophageal squamous cell carcinoma.Results:1.Immunohistochemical test results showed that the positive expression rate of ABCE1 protein in esophageal squamous cell carcinoma tissues was 72.92%(35/48),while the positive expression rate of ABCE1 protein in adjacent normal esophageal tissues was 35.42%(17/48).There was a difference between the two(P<0.05).2.The results of Western Blotting(WB)showed that the expression level of ABCE1 protein in esophageal squamous cell carcinoma tissues(0.955±0.228)was higher than that in adjacent tissues(0.343±0.163)(P<0.05).Phase 3.Statistical analysis showed: ?ABCE1 protein expression in esophageal squamous carcinoma tissues levels(0.662 + 0.074)higher than that of patients with stage ?,expression level(0.386 + 0.023)(P < 0.05);?The expression level of ABCE1 protein in low-differentiated esophageal squamous cell carcinoma(0.953±0.084)was higher than that in high-medium-differentiated esophageal squamous cell carcinoma(0.632±0.032)(P<0.05).The expression level of ABCE1 protein in lymph node positive patients(0.580±0.085)was higher than that in lymph node negative patients(0.326±0.042)(P<0.05).There was no significant difference in ABCE1 protein expression between ?60 years old(1.267±0.023)and <60 years old(1.189±0.016)(P>,0.05).There was no significant difference in the expression level of ABCE1 protein between male patients(1.366±0.026)and female patients(1.287±0.033)(P>,0.05).There was no significant difference between ABCE1 protein expression level(1.280±0.357)and ABCE1 protein expression level(1.134±0.463)in patients who drank alcohol(P>,0.05).Conclusion:The positive expression rate and expression level of ABCE1 protein in esophageal squamous cell carcinoma tissues were higher than those in adjacent tissues.The expression level of ABCE1 protein in patients was correlated with tumor staging,differentiation and lymph node metastasis,and had no relation with age,gender or alcohol consumption.The experimental results show that ABCE1 may become a new research direction for the study of the occurrence and development of esophageal squamous cell carcinoma,providing a new research idea for clinical biotherapy.
