ObjeetivesTo observe the relevance of platelet P2Y12 gene C34T, G52T single nucleotide polymorphisms and ischemic stroke.MethodsThe case group selected 169 of unrelated patients with ischemic stroke, in which male 95 cases, female 74 cases, with an average age of 64.51±10.6 years old. Control group,186 patients, including 101 male and 85 female, with an average age of 62.67±6.49 years old. Determination the clinical indication associated with CIS,including blood fat,blood glucose,high blood pressure and BMI. Subjects fasted 12 hours later, the patients were admitted the next morning, volunteers were on the examination day, collecting peripheral venous blood 2ml, with EDTA anticoagulant. Use DNA extraction kit to extract genomic DNA, and conducted DNA quantification and purity analysis.To detect the mutation genotype and allele frequencies of platelet P2Y12 gene C34T, G52T with PCR-RFLP. Comparison the relationship of two groups of genotype and allele frequency distribution and with ischemic stroke.Results1. The platelet P2Y12 gene C34T mutation allele frequency in Chinese populations and patients with ischemic stroke were21.51% and23.67%, mutation frequency in Group has no significant difference in population (P> 0.05).2. The platelet P2Y12 gene C34T, G52T mutation allele frequency in Chinese populations and patients with ischemic stroke were 18.82% and 22.78%, mutation frequency in Group has no significant difference in population (P> 0.05). 3. The case group C34T sites CC, CT, TT genotype frequencies were 59.17%, 34.32% and 6.51%; the control group C34T sites CC, CT, TT genotype frequencies were 62.37%,32.26% and 5.37%, two sets of genotype frequencies consistent with Hardy-Weinberg equilibrium, but the genotype frequencies in the two groups has no significant difference in the distribution of population (P> 0.05).4. The case group G52T sites GG, GT, TT genotype frequencies were 60.95%, 32.54% and 6.51%; the control group G52T sites GG, GT, TT genotype frequencies were 67.20%,27.96% and 4.84%, two sets of genotype frequencies consistent with Hardy-Weinberg equilibrium, but the genotype frequencies in the two groups has no significant difference in the distribution of population (P>0.05).ConclusionThere is no significant difference between the C34T, G52T single nucleotide polymorphism of the platelet P2Y12 gene in Chinese healthy people and patients with ischemic stroke.
