IntroductionObesity is a kind of endocrine metabolic disease characterized by excessive body fat deposition and distribution.Is a kind of disease caused by appetite and energy regulation disordered, it is related with genetic, environmental, dietary patterns and other factors, its causes and genetics, central nervous system abnormalities, nutrition, endocrine disorders and other factors are closely related.Obesity is not only leading to cardio-cerebral vascular disease risk factors, increased diabetes, hypertension, hyperlipidemia, fatty liver, as well as 15 kinds of diseases such as cancer incidence, but also brought many social problems and psychological problems.However, there is a long-term study of the mechanism of obesity stagnating, medication made little headway, In the past few decades, obesity has been proved that many endocrine, neurological, hormonal, cellular and other ways to offset.In recent years, found that many biological factors is related with the pathogenesis of obesity, such as free fatty acids, glycerol, hormones, inflammatory factors and so on. Understand the role of cytokines and related effects, is important to the prevention and treatment of obesity and insulin resistance.1998 Orexin (orexin) discovery caused people to have some new understanding of obesity-related hormones and neuropeptides, as well as the mechanisms of obesity, orexin become a hot research for the current diet-induced obesity, provides a new way to the treatment of obesity.At home and abroad, there are few studies on the relationship of orexin receptor and nutrition obesity.Therefore, this topic will identify priorities for research for the orexin receptorl in the obese rat hypothalamic expression and the correlation between orexin receptor land obesity, provide theoretical basis for the incidence and Prevention of this metabolic syndrome core elementObjectiveTo investigate the expression of orexin receptor1 in Obese rat hypothalamus and to reseach the correlation between orexin receptor land obesityMaterial and methodsTo establish and evaluate a high-fat diet obese rat models, Lateral ventricular cannulization was performed in obese rat which then received an intracerebroventricular injection of orexin receptor antisense oligodeoxynucleotide, blood glucose were measured by blood glucose meter, the levels of blood fat were measured by using biochemical color matching technique and enzyme technique, and insulin were measured by chemiluminescent immunoassay, expression of orexin receptor 1 were analyze by immunohistochemical.ResultRats were raised by high-fat diet eight weeks,the weight,Lee's indexes,blood glucose,insulin,TC,TG and LDL-C of obese group showed significantly higher when compared with control group.(P< 0.01). Expression of orexin receptorl in rat hypothalamus tissue showed significantly higher when compared with control groups(P< 0.01). After the obese rats were treated with orexin receptorlantisense oligodeoxynucleotide 48h, the expression of orexin receptorl in rat hypothalamus tissue and the levels of blood glucose, insulin in the antisense group were markedly lower than those in the saline group, and the mismatched group. (P< 0.05)Conclusions1.The high fat diet can induce obesity to rats.2. OXRlis expressed in the cytoplasm of the hypothalamus neurons, In the process of pathology nutritional obesity, rats OXR1 protein may participate in the hypothalamus energy metabolism regulation.
