Shuangbai powder is empirical formula that widely used in surgery and Orthopedics, is made up of Radix et Rhizoma Rhei,Platycladus orientalise,Cortex Phellodendri Amurensis,Lycopus and Mentha, with the efficacy of activating blood and removing stasis, detoxify, relieve swelling and pain. It has some disadvantages such as backward process,wasting medicinal materials,inconvenient tobe used,easy to stain clothing. The research purify effective ingredients of Radix et Rhizoma Rhei and Cortex Phellodendri Amurensis of Shuangbai powder, aimed to lay the scientific proof for developing determined curative effect,convenient to be used external use spraying film of TCM.Aim:Optimize the technology of Radix et Rhizoma Rhei and Cortex Phellodendri Amurensis and study on forming process of Shuangbai powder. The research aimed to lay the foundation for developing determined curative effect,convenient to be used external use spraying film of TCM.Methods:(1) With the contents of aloe-emodin, rhein, Rheum emodin, chrysopHanol and pHyscion as indexes, contents determination of alcohol extracts by HPLC, ascertain the optimum extraction and purification technology, Purifying aloe-emodin, rhein, Rheum emodin, chrysopHanol and pHyscion from Radix et Rhizoma Rhei by D301 macroporus resin, study on different,different concentration,the leakage rate of the sample solution, the acid and volume of eluent. (2) Purifying the alkaloids from Cortex Phellodendri Amurensis by AB-8 macroporus resin, With the contents of berberine hydrochloride and palmatine as an index, contents determination of alcohol extracts,the sample solution and eluent from AB-8 macroporus resin were done by HPLC. (3) Do forming experiments with polyvinyl alcohol (PVA),chitin,carbomer,polyvinylpyrrolidone (PVP),Chitosan and carboxymethyl cellulose sodium (CMC-Na), with spray experiment,forming time,dynamic viscosity,configuration of film surface as indexes, do preliminary screening on forming accessory of spraying film. (4) Observe the configuration of surface of each test articles after forming by bio-microscope, study on the forming affection of five purificatory materials of Shuangbai powder (anthraquinone of Radix et Rhizoma Rheiand,flavonoids of Cacumen Platycladi,alkaloid of Cortex Phellodendri Amurensis,flavonoids of Lycopus,volatile oil of Mentha) on forming accessory, ascertain the optimum forming accessory of spraying film.Results:(1) The optimum technology of Radix et Rhizoma Rhei were:using 5 times 95%(V/V) ethanol as extractor and extracting for 1h and 3 times, the transfer rate of Total anthraquinones was 98.34%, the transfer rate of Total anthraquinones was about 90% when the concentration of sample solution was 0.1g·mL-1 and pH was 11. The best eluent was 20 times 0.25mol·L-1 HCL 75% ethanol and the transfer rate of Total anthraquinones was about 75%. (2) The optimum technology of Cortex Phellodendri Amurensis were:using 8 times 75%(V/V) ethanol as extractor and extracting for 1h and 3 times, the transfer rate of Alkaloidswas 83.75%, the transfer rate of Alkaloids was about 80% when the concentration of sample solution (adjust pH 8 and centrifugation) was 0.1g·mL-1, the transfer rate of Alkaloids in eluent was 75.14% when sample solution was 0.1g·mL-1。(3) Chitosan and chitin are difficult to dissolve in water and ethanol in the case of room temperature or heated; Carbomer soaked in water overnight, dissolved gel was difficult to eject, plug the nozzle easily; CMC-Na has good solubility, but it can not forming, so they are not suitable as film forming material of spraying film; PVP have good forming, and proportional relationship between its molecular weight, owing to the molecular weight of PVP K15, K17, K25 smaller than PVP K30, the amount of PVP K15, K17, are less than K30 to achieve the same effects, however, PVP K90 has large molecular weight, it is difficult to eject when dissolved, and easy to flow away after ejecting. Chosing PVP K30 base on cost, and forming experiments show that it has good forming when the concentration between 6%-10%; PVA have good forming, but it like linear when the concentration is high, so choose a critical concentration of their linear injection experiments, PVA-124 is 1.2%, PVA-1788 is 1.6%, the low viscosity type of PVA-1788 is 2.4%(4) Although PVA-124,PVA-1788,PVA-1788 low viscosity have good forming effects on the purificatory materials of Cacumen Platycladi,Cortex Phellodendri Amurensis,Lycopus, Mentha, from which the spraying film is made with purificatory materials of Radix et Rhizoma Rheiand or the mixture of five purificatory materials from Shuangbai powder is muddy, after forming have obvious deposition, the film transparence,dense, no obvious deposition, and PVP K30 is apt to dissolve in water and ethanol. It don not need heating when dissolution, handle easy, as the best accessories.Conclusion:Ascertain the purification technologies of Radix et Rhizoma Rhei and Cortex Phellodendri Amurensis, study on the film-forming property of various forming accessories, study on the common technology of forming between purificatory materials of Shuangbai powder and forming accessories for the first time, ascertain polyvinylpyrrolidone (PVP) K30 as the best accessories of spraying film, lay the foundation for developing Shuangbai powder.
