Part 1Relationship between expression of apoptosis inducing factor and neuronal apoptosis during focal reversible ischimia in rat brainObjective:To study the relationship between expression of apoptosis inducing factor and neuronal apoptosis in ischemic penumbra when rat focal ischemia reperfusion after focal middle cerebral artery occlusion established.Methods:Wistar rat middle cerebral artery occlusion (MCAO) reperfusion model was established by thread ligation. AIF expression in ischemic penumbra and nerve cell apoptosis degree in sham operation group, MCAO 2 hours and reperfusion 6h, 24h,48h,72h and 168h were observed by TUNEL and immunohistochemistry techniquel.Result:AIF increased since 6h after ischemic reperfusion and peaked at 48h of reperfusion in the ischemic penumbra. Cell apoptosis occured in all groups of ischemic reperfusion, the percentage of apoptosis cells in 48h reperfusion group was the highest among all of the groups. There were significant difference among all groups (P<0.05)Conclusion:I/R could induce the increased AIF positive cell expression and increase neuronal apoptosis.Partâ…¡Effects of ifenprodil on expression of AIF and neural cells apoptosis in rat cerebral ischemia-reperfusionObjective:To investigate the effects of ifenprodil on expression of AIF and neural cells apoptosis in rats cerebral ischemia-reperfusion.Methods:Wistar rats middle cerebral artery occlusion reperfusion model was established by thread ligation. Rats were observed by neurological deficit score, TTC staining, HE staining, Nissl staining, TUNEL and immunohistochemical staining in sham operation group, the control groups and Ifenprodil intervention groups (10mg/kg, ip) about performance and expression on the same time points.Result:Ifenprodil therapy can reduce the rate of rat nerve function, reduce infarct size, reduce edema, increase the neuronal survival,reduce AIF-positive cells and apoptosis. Compared with control group, the part of the data has statistical difference (P<0.05).Conclusion:Ifenprodil can reduce neural cells apoptosis through inhibition of NMDA receptor NR2B subunit, decrease EAA toxicity and expression of AIF,and to inhibite the apoptosis induced by AIRPartâ…¢Influence of Danhong injection on the AIF expression and neuronal apoptosis in cerebral ischemia-reperfusion ratObjective:To study the Influence of Danhong injection on the AIF expression and neuronal apoptosis changes in cerebral ischemia-reperfusion rat.Methods:42 of Wistar rats were randomly divided into:sham-operated group, focal cerebral ischemia-reperfusion group (control group), and Dan Hong injection treated group (treatment group), control group and treatment group were further divided into 1d,3d,5d groups. The establishment of a modified suture method in rats with focal cerebral ischemia-reperfusion model, in each group of the control group and treatment group, respectively take brain tissue on 1d,3d,5d,measured AIF and neuronal apoptotic changes in cerebral ischemic penumbra.Results:The expression of AIF and neuronal apoptosis in treatment groups on 1d,3d, 5d were significantly lower than the control groups, compared with the control groups,it has a statistically significant difference (P<0.05).Conclusion:The Danhong injection can inhibit the expression of AIF,reduce neuronal apoptosis and has cerebral protective effect.
