Background and PurposeCerebrovascular disease is one of the common diseases which is a serious threat to human health of the elderly, with high incidence, high morbidity and high mortality, and more than80%of which is ischemic cerebrovascular disease. Ischemia-reperfusion injury is more serious brain injury which is defined as tissue damage caused when blood supply is restored to the ischemic infarct brain regions after a period of ischemia,which causes further damage of the lesion tissue.Inhibition of reperfusion injury is a key link in the treatment of ischemic cerebrovascular disease. Although domestic and foreign researchers have conduct a lot of research and have made some progress,there is still lack of targeted treatment strategies.Chinese medicine treatment of ischemic cerebrovascular disease has a long history, with the wide range of advantages such as strong side effects, pharmacological effects, which can play a therapeutic on multiple levels and multiple links, particularly suitable for a variety of pathological changes induced by cerebral ischemia.in the long-term practice,the classical theory of cerebral ischemia, herbs has been attract the attention by the medical prefession.According to the several years clinical experiences. Professor Li Ping has formed an effective prescription-HuoXueKaiQiao-decoction. It is a mixture of some herbs, such as Tianma, kudzu root, Chuanxiong, lumbricus, Salvia, Polygala, Achyranthes, red peony root, mistletoe etc. With the effectiveness of tonifying kidney and promoting blood circulation, clinical practice shows that this pharmacy received significant clinical effect in the treatment of ischemic stroke. This study by observing the changes of NO content and the number of neuronal apoptosis in rat brain tissue to explore the mechanism of HuoXueKaiQiao-decoction on cerebral ischemia-reperfus-ion injury and supply further theoretical and experimental basis for clinical medication in order to promote application.MethodsIn the subject, to test the the neuroprotective effects of the Chinese medicine KaiQiao HuoXue-decoction,adult male Wistar rats were used as experimental animals. Male Wistar rats were randomly divided into5groups:blank group, sham group, model group and therapy group. Every group are breed in different cages under the same condition. Rats of the therapy group were intragastrically administered the appropriate doses of Chinese medicine HuoXueKaiQiao-decoction,and the same doses of Physiological Saline to the sham and model groups each day for5days before ischemia induction, and administration was continued until sacrifice at conclusion of the experiment. The cerebral ischemia was induced by90min of middle cerebral artery occlusion (MCAO), which was based on the method described initially by Zea-Longa with improvement. At24hours and48hours after the models were made, we rapidly kill the rats and completely take out of the brain tissue,half of which are made to homogenate to detect the NO content.The other brain are taken after heart perfusion by paraformaldehyde, and to be used to observe the Apoptosis and pathological change by the way of staining. We detect the content of NO in the brain tissue fluid samples by using nitrate reductase method in strict accordance with the steps of the kit, observe the apoptosis by TUNEL.and pathological change by hematoxylin and eosin stain.ResultsBy experimental observation, pathological observations indicate that after24h,48h the number of the damaged nerve cell of the rats that treated by HuoXueKaiQiao-decoction was large, and damaged severely. While the degree of damage are significantly lighter in the therapy group. It was determined that the content of NO and the neuronal apoptosis in the brain tissue of rats in model group were significantly increased (P<0.01), while the NO content of brain tissue in the therapy group rats decreased significantly compared with the model group (P<0.05), and apoptotic cells decreased significantly too.(P<0.01),ConclusionsHuoXueKaiQiao-decoction can improve the nerve function of the rats with ischemia-reperfusion injured24h and48hours and mitigate the damage to the brain tissue by reducing the content of NO,the experimental results also show that the HuoXueKaiQiao-decoction can inhibit the development of ischemic cells apoptosis, delay or prevent the neuronal death process of ischemic area,to some extent,to salvage damaged neuron sand prevent the expansion of infarction lesions.
