Objective:To compare the expression of TFF2,CDX2 and PTEN in gastric cardia, and to explore the relationship betwwen the cardia metaplastic lesions and gastric cardia carcinogenesis. Methods:A total of 79 specimens (39 gastric cardia adenocarcinoma and 40 antral carcinoma) were analyzed, the expression of TFF2,CDX2 and PTEN were determined by immunohistochemical methods of two-step.Results:1. The expression rates of TFF2 were 33.3% and 10.0% in GCA and AC, and in adjacent tissues were 63.3% and 30.3%, respectively (p<0.05 and p<0.01).2. The expression rates of CDX2 were 20.5% and 70.0% in GCA and AC (p<0.01), and in adjacent tissues were 13.3% and 42.4%, respectively (p<0.05).3. The rates of intestinal metaplasia were 41.0% and 72.5% in GCA and AC respectively (p<0.01). 4. The both positive rate of CDX2 and PTEN was 65.0%(p<0.01) in antral carcinoma, the consistency of CDX2 and PTEN is general.Conclusions:1. The gastric cardia adenocarcinoma may be related with the cardia metaplasia lesions, and it was more closely related with spasmolytic polypeptide(SP/TFF2)-expressing metaplasia(SPEM).2. The combination determine of TFF2 and CDX2 can predict the risk of early gastric cardia adenocarcinoma.3. The CDX2 and PTEN play a synergistic role in antral carcinogenesis, and combination determine of CDX2 and PTEN can predict the risk of antral carcinoma.
