Aim: Using microarray technology,the lncRNA and miRNA in gastric cardia adenocarcinoma(GCA)tissue and para-carcinoma tissue were detected.The aim of this study was to study expression profiles and correlation of long non-coding RNA(lncRNA)and micro RNA(miRNA)in GCA and analyze the association of abnormal mi RNA expression and prognosis.Materials and methods: A total of 52 cases of GCA tissue and para-carcinoma tissue excised in surgery were researched.Total RNAs were extracted and detected in15 GCA specimens and PCR was performed in the other 37 specimens.The expression profiles of lnc RNAs and mi RNAs in GCA were detected by Arraystar Human Lnc RNA microarray V2.0 and miRCURY LNA? microRNA Array,7th gen-hsa,mmu & rno.The differentially expressed lncRNAs and miRNAs were obtained by RNA labeling,array hybridization and data analysis in 15 cases of GCA tissue and para-carcinoma tissue.The association with the expression of lncRNA and miRNA was analyzed by bioinformatics prediction approach.QRT-PCR was used to verify the above results in 37 cases of GCA tissue and para-carcinoma tissue.The correlation of mi RNA expression level and clinicopathologic characteristics was analyzed by Chisquared test.Kaplan-meier survival analysis was used to analyze abnormal miRNA expression and survival.Results: 2090 lncRNAs,152 miRNAs(fold change ? 2)were detected in 15 cases of GCA tissue and para-carcinoma tissue.LncRNA H19 and MALAT1 were selected from lncRNA expression profiles based on multiple of differential expression,values of normalization intensity and annotation of lnc RNA databases.According to bioinformatics prediction approach,three miRNAs including has-miR-148a-3p,hsami R-205-5p and hsa-miR-363-3p are associated with H19 and MALAT1 for GCA.Upregulated H19 and down-regulated miR-148a-3p were verified by QRT-PCR in 37 cases of GCA tissue and para-carcinoma tissue.The result of PCR is consistent with the microarray analysis.The correlation between H19 and miR-148a-3p was observed by nonparametric rank sum test(P = 0.019).Down-regulated expression of mi R-148a-3p is significantly associated with lymph nodes metastasis(LNM)(P =0.039)and survival(P = 0.022)for GCA patients based on clinicopathologic characteristics and kaplan-meier survival curve.Conclusion: Comparing with para-carcinoma tissue,GCA tissue contains a lot of differentially expressed lncRNAs and miRNAs.The correlation of H19 and seven mi RNAs was detected based on bioinformatics prediction approach.Moreover,low expression of mi R-148a-3p is linked to survival and LNM for GCA patients.
