The Polygonum Perfoliatum L. is one pecies of Polygonaceae, and which is used in folk medicine. The whole plant of Polygonum Perfoliatum L. is widely distributed in China, the plant was used as folk medicine in treating edema, jaundice, dysentery, whooping cough and so on. Recently, some studies showed that the Polygonum Perfoliatum L. has anticancer activity, and less in research of the plant. In orde to investigate the bioactivities compounds, Polygonum Perfoliatum L. was made a systematic study, eight compounds were isolated, purified from the plant. Their structures were determined by 1D and 2D NMR analysis, respectively, one of them is new compound, two are first isolated from this plant, and some of them were did preliminary research. The results are as follows:1. The chemical constituent in the petroleum ether extract of Polygonum Perfoliatum L. was separated by repeated chromatography on silica gel to obstain compound P-Sitosterol(Ⅰ).2. The chemical constituent in the ethyl acetate extract of Polygonum Perfoliatum L. was isolated by chromatography on silica gel, Sephadex LH-20, RP-18 and Pre-HPLC, and gain four compounds: Pentacosanoic acid(Ⅱ),3,4-Dihydroxybenzoic acid(Ⅲ), Quercetin(Ⅳ), 8-oxo-pinoresinol(Ⅴ). CompoundⅤis new compound.3. The chemical constituent in the n-butanol extract of Polygonum Perfoliatum L. was purified by D101 macroporous adsorption resin eluted with water,25%,50%,75% and 95%ethanol to obtain the components with different polarity. The fraction of 50% ethanol was isolated by sephadex LH-20 to obtain compounds quercetin-3-O-β-D-glucuronide (VI) and quercetin-4'-O-β-D-glucuronide (Ⅶ); The fraction of 75% ethanol was isolated by chromatography on silica gel, sephadex LH-20 to obtain compound Rutin(Ⅷ). CompoundsⅡandⅦwere first isolated from this plants.4. Biological activity of compoundsThe research discovered the flavonoids of Polygonum Perfoliatum L. have strong antioxidant activity. The DPPH free radical-scavenging of the flavonoids up to 80% under low concentration about 25.0μg/mL. And what is more, the structure-activity relationship of flavonoids antioxidant was discussed.In vitro test (MTT method), compound (Ⅳ) showed moderate anti-tumor activities against human prostate carcinoma(PC3). Compound V showed that inhibition effects on human hepatocellular carcinoma (SMMC7721) more better than human prostate carcinoma(PC3).In addition, the advances about components from Polygonaceae were reviewed. The results of present study will be useful to further phytochemical investigation on Polygonum Perfoliatum L.
