The Polygonum Perfoliatum L. which also known as Guan-Ye-Liao, She-Dao-Tui, is one pecies of Polygonaceae. It’s distribute mainly all over our country which used as folk medicine in treating edema, dysentery, whooping cough, jaundice and so on. In recent years, many attention have taken to the Polygonum Perfoliatum L. by some famous scholar, and what’s more, it’s recorded in2010Chinese Phamacopoeia. The chemical composition is very complex which contains Flavones, Quinones, Tepenoids, Phenol acids, Phenylpropanoid sugar esters and so on. Not only the chemical composition is very complex, but also it contains wide pharmacological effects, such as anti-inflammatory, anti-virus, anti-tumor and so on.This dissertation consists three sections. The first section introduction the compounds isolated and purified from The Polygonum Perfoliatum L. In the second section, we establish a new method to determinate the quercetin and the total flavonoids of the Polygonum Perfoliatum L. by UV-Vis spectrophotometry and RP-HPLC. at the last section, we presents an overview about domestic and oversea researches on main chemical constituents and pharmacological effects of Polygonum Perfoliatum.L, so that to provide with a basis for its development, utilization and further research.The chemical constituents of Polygonum perfoliatum L. were isolated and purified by chromatography on silica gel, Sephadex LH-20and Semi-preparative HPLC. The structures of compounds were elucidated and identified by physicochemical properties,1H-NMR,13C-NMR and ESI-MS. Ten compounds were isolated and identified as5-Hydroxymethyl2-furald Ehyde (1), Methyl caffeoate (2), Protocatechuic aldehyde (3), Quercetin (4), Pinocernbrin (5), catechin (6), Taxifolin (7), Taxifolin-3-O-β-D-xylopyranoside (8),13-Epitorulosal (9), Coumarin-7-O-β-D-glucose glycosidic (10). Among them, Compounds6.7、8、9were isolated from the plant for the first time.The determination contents is determinated by UV-Vis spectrophotometry and RP-HPLC. The result shows that there is no significant difference between this methods and Chinese Phamacopoeia of the contents of the total flavonoids. But in the other side, the contents of quercetin is very obvious. At last, We use the same methods to determinate the contents of the total flavonoids and the quercetin of the sample which refined by the D101Macroporous resin, the result shows the contents of the total flavonoids in the refined sample is more than ten times than the previous sample.
