Objective:To observe the difference of protective effect between FTY720 and Methylprednisolone after spinal cord injury.Method:224 female SD rats were randomly divided into A,B,C,D 4 groups,56 rats in every group. Rats SCI were induced using Allen's T10 spinal cord injury model.A group(sham group):cut theT9,T10 lamina only,no spinal cord injury, feeding 0.3ml saline, received a tail vein injection of 0.3ml saline;B group(comparison group):feeding 0.3ml saline and received a tail vein injection of 0.3ml saline after spinal cord injury;C group(methylprednisolone group):rats received a tail vein injection of methylprednisolone (30mg/kg in 0.3ml saline) after spinal cord injury,and feeding 0.3ml saline;D group(FTY720 group):feeding FTY720(3mg/kg in 0.3ml saline)and received a tail vein injection of 0.3ml saline after spinal cord injury. Execute rats to take injury tissue in 6h,12h,24h,48h,72h,1w,3w after surgery,take 8 in every group every point.Take HE staining,Immunohistochemistry staining,Tunel staining,and observe spinal cord necrosis,inflammatory cell infiltration,capase-3 expression,apoptosis and so on.Take behavior observation(BBB Score and Inclined Plane Test) on 1d,3d,7d,14d,21d after surgery,and detect cortex somatosensory evoked potential on 21d after surgery.Result:Morphology and function of the spinal cord are close to normal in A group. Spinal cord necrosis and functional recocery,the worst in B group,the second in C group,the best in D group.The results of Inclined Plane Test,BBB Score and cortex somatosensory evoked potential,D group is better than C group, D group and C group are better than B group,There are different significant (P<0.05). Conclusion:Both FTY720 and methylprednisolone have protective effect after acute spinal cord injury in rats,they also can reduce secondary spinal cord injury, but from the treatment effect, FTY720 are better than methylprednisolone.
