Objetives Establish an animal model of PTE by FeCl3 aqueous solution injection into the sensorimotor cortex of rat, observe characteristics of epilepsy seizures and pathology changes in rat brain after administration of Lamotrigine at different time and dosage.Materials and methods 84 adult male Sprague-Dawley rats were randomly divided into 7 groups, namely normal control group, model control group, low, middle and high-dose LTG prevention group, low, mid-dose LTG treatment group. In each group, acute-onset latency, frequency of seizures was observed, acute and chronic EEG by cortical electrodes was recorded, neuron loss, gliosis and dentate gyrus mossy fibre sprouting in ipsilateral hippocampi was studied by Nissl staining,GFAP immunohistochemical analysis and Timm staining,respectively.Results After injection of FeCl3, EEG shows paroxysmal or sustained release of epileptic waves after a latency of 376.14±18.78s,the most highest amplitude is up to 600μv. At the same time behavioral changes of epilepsy are observed and the Racine grade level is up to 3-5; Latencies in prophylactic LTG administration groups are all prolonged compared with model control groups and therapeutic LTG administration groups,while latencies in mid-dose prevention group and high-dose prevention group are of no significant difference.The total number of SRS in 7-14d after modeling in LTG administration groups is small than model group. The number of SRS in prophylactic LTG groups is fewer than therapeutic LTG groups, but there is no significant difference in mid-dose LTG prophylactic and therapeutic administration groups. Pathology study of ipsilateral hippocampi 2 weeks after modeling reveals:neuron loss in model group is more remarkable (26.29±1.11)than in normal group(51.75±2.37), Gliosis in model group(44.43±2.70) is more than in normal group(28.50±1.20),there is remarkable differdnce in low-dose, mid-dose LTG prophylactic administration groups and the same dose LTG therapeutic administration groups. Scale of MFS in dentate gyrus of model groups(2.00±0.27) is significantly reduced by LTG administration and there is no difference in each prophylactic LTG administration group.Conclusions (1)Prophylactic administration of LTG prevent EP by prolong incubation period,reduce SRS and improve EEG. Prophylactic administration has more obvious effects than therapeutic administration in reducing SRS. (2) Both prophylactic administration and therapeutic administration of LTG prevent nerve cell from death obviously, gliosis is the same. Prophylactic administration has more obvious effects than therapeutic administration.(3) Both prophylactic administration and therapeutic administration of LTG reduce MFS in dentate gyrus, there is no difference in LTG administration groups.(4)Mid-dose and high-dose LTG prophylactic administration have similar effects in EP prevention, high-dose LTG administration exhibits side effects.
