Objective1.By testing the expression of P16,Ki-67 in different cervical tissues and understanding the relationship of P16,Ki-67 in different levels of cervical intraepithelial neoplasia.2.To investigate the role of P16,Ki-67 double staining in diagnosis and grade of cervical intraepithelial lesions.Materials and methods1.137 cases was selected in the first affiliated Hospital of Zhengzhou University from January 2011 to December 2015,by HE staining pathological diagnosis in the hospital to detect 15 cases of immature metaplasia(IM),32 cases of low-grade squamous intraepithelial lesion(LSIL),90 cases of high-grade squamous intraepithelial lesion(HSIL).2.The staining of P16 and Ki-67 in all sections was detected by immunohistochemical staining SP method,and then detected the co-expression of P16 and Ki-67 by immunohistochemical double staining in all tissues.3.Statistical analysis: The SPSS 17.0 statistical software package was used for data analysis and ?2 text.With P<0.05 considered to represent a statistically significant difference.Results1.P16 was negative in IM and being focal or patchy staining in LSIL with weak intensity.While,the positive cells were restricted to the lower third of the cervical epithelium.In HSIL,diffuse P16 immunostaining of two-thirds or full thickness positivity of the cervical epithelium.The positive rates of P16 protein were 0%,62.50%,92.22% in IM,LSIL and HSIL respectively,and differences between these three groups were statistically significant.2.For the IM,Ki-67 expression only existed in the basal or parabasal cells.In LSIL,Ki-67 immunostaining restricted to the lower third of the cervical epithelium.While,diffuse Ki-67 immunostaining of two-thirds of the cervical epithelium or full thickness was seen in HSIL.The Ki-67 index was 7.21%,16.41%,43.46% in IM,LSIL and HSIL respectively,and differences between these three groups were statistically significant.3.The dual staining does not lead to the absence of positive signal.Ki-67 index was 35.14% in P16 positive region,while was 7.68% in P16 negative region.With the increase in classification of cervical lesions the cases of P16 and Ki-67 co-expression increased.The co-expression rates of P16 and Ki-67 were 0%,51.35%,96.04% in IM,LSIL and HSIL respectively,and differences between these three groups were statistically significant.ConclusionsThe expression of P16 and Ki-67 is associated with grade of cervical intraepithelial lesions.There is a significantly positive correlation between the expression of P16 and Ki-67.Immunohistochemical dual stain is helpful to diagnosis and grading of cervical intraepithelial lesions.
