Objective:To research the pharmacognosy features and establish the fingerprints of two kinds of mistletoe, and screened its pharmacodynamic activity of the 50% CH3OH extracts of them.Methods:Experiment 1:The transaction of the stalk part of two kinds of mistletoe and powder film were made using the conventional method for further researching on anatomy.Experiment 2:Fingerprints were established by the method of TLC, HPLC and RAPD.Experiment 3:The Human ovarian cancer cell line HO-8910, Human gastric cancer cell line SGC-7901 and zebrafish inhabiting angiogerlesis model were used for screening the anti-tumor activities of the 50% CH3OH extracts of the two kinds of mistletoe.Results:Experiment 1:Viscum articulatum Burm and Viscum coloratum (Kom.)Nakai could be distinguished by the transaction and powder film.Experiment 2:The toluene-ethyl acetate-glacial acetic acid (8:2:0.1) mobile phase was used to establish the TLC fingerprints, and all the samples contained the Coca-diol,β-incense resin, a mixture of alcohol and lupeol, and oleanolic acid; The gradient elution method was used to get the HPLC fingerprints of two kinds of Mistletoe and their similarities were all above 0.7; The low pH value, high-salt method was used to extract the genomic DNAs of medicinal herbs, and after agarose gel electrophoresis,7 pieces of random primers from 25 (S1-S25) can amplify the polymorphism of the strip, and the amplify effect of S16 to two kinds of Mistletoe was well.Experiment 3:In the MTT experiment, the IC50 ofⅤ. coloratum (Kom.)Nakai on HO-8910 and SGC-7901 were 1.85-8.85 mg/mL,1.91-7.00 mg/mL, and the IC50 ofⅤ. articulatum Burm were 0.64-1.97mg/mL,0.38-1.25mg/mL; Concerning the zebrafish experiment, the extracts of V. coloratum (Kom.)Nakai had no anti-tumor activity at the concentration of 10μg/mL and 100μg/mL, but the extracts of V. articulatum Burm had a favorable action, the inhibiting rates were 41.9-100% at the concentration of 100μg/mL and 28.5-35.87% at the concentration of 10μg/mL.Conclusions:In the experiment, the ecology research, the fingerprints of TLC, HPLC, RAPD were established, which providing theoretical basis for the identification, standardization and quality standards of the two kinds of mistletoe. And the pharmacodynamic research were the base for drug research and development and utilization, which having the value of further study.
