Objective:The osteoblastic cells were cultured in vitro and taking three intervention measures respectively,including ESW,PDGFand ESWplusPDGF,oberving the proliferation and differentiation of the cells, MTT assay was employed to observe the proliferation of osteoblasts, supernatant fluid were detected by the method of ELISA to contain the quantity of BMP-2 and discuss the effect for extracoporeal shock wave plus PDGF on proliferation of osteoplast and promote the secretion for BMP-2.Methods:The osteoblastic cells were cultured and prepare cell suspension.According to the methods of different intervetion,four experimental groups were entered[ESW (A) PDGF (B) ESW plus PDGF (C) and black group (D)];After relevant intervetion,at the temperature 37℃,5%CO2,the osteoblastic cells were cultured for 24h,48h,72h in the thermostaten。oberving the proliferation and differentiation of the cells in microscope,MTT assay was employed to observe the proliferation of osteoblasts, supernatant fluid were detected by the method of ELISA to contain the quantity of BMP-2.Results:Observing the cell and extracting cell suspension fluid each time,the cell were counted in the slides Through the MTT,we could discover the phenomenon,in the four experimental group,the group(C) is the most active,the group(D) is the least active,especially at 48h。Comparing with group (A) (B) (D), group (C) is different from the others.(p<0.01) At the same time,BMP-2 in the insulution fluid,the group (C) is more concentrated than any other, the difference is existed among the four groups.(p<0.05).Conclusion:ESW,PDGF could stimulate the proliferation of osteoplast,but combining the two intervetion could enhance the action.
