Background Irritable bowel syndrome is a kind of abdominal pain or discomfort with the change of defecate habit,and there was no histology change or biochemistry abnormal,belong to intestinal tract funtion disorder disease. Neal et al first put forward the conception of PI-IBS . Guomin et al made the post-infective intestinal function disorder rat model was induced by Shigella flexneri intragastric, imitated the course of human PI-IBS development. There were study indicated that, part intestinal inflammation and immunoregulation abnormal may participate in the course of PI-IBS development. Dendritic cells which was the main antigen presenting cells was more attentioned by people in the role of initiating intestinal mucoal immune reaction. This study through established the rat model of post-infective intestinal function disorder, observed the expression of rat intestinal dendritic cells and related cytokines, discussed the function change of rat intestinal immune tolerance .Objective Through established the rat model of post-infective intestinal function disorder, this study observed the mature and expression of rat intestinal dendritic cells, and detected interleukin-1β(IL-1β) and interleukin-4(IL-4) expression of rat peripheral blood and intestinal, compared the difference between ileum and colon, discussed the function change of rat intestinal immune tolerance.Methods The 60 male Wistar rats were randomly divided into control group and experimental group, the rat model which intestinal function disorder after infection was induced by Shigella flexneri intragastric. Shigella flexneri 1 ml in a concentration of 9×108CFU/ml was injected into 30 Wistar rats of experimental group by gavage to make models of post-infectious bowel dysfunction , and equivalent saline into the other 30 rats of normal control group by gavage. Stool form and histological changes of distal colon, cultivated shigella flexneri were observed at day 1 , 4 , 7 , 10 , 13 and the perceptual threshold was detected with colon-rectal balloon distention at day 16 ,19 , 22 , 25 after infection. After the model established, evaluated the macroscopic and histological scores of terminal ileum and distant colon of two groups, applied immunohistochemistry to detect the expression of intestinal surface molecules CD11c and constimulatory molecules CD80 and CD86, and detected the expression of interleukin-1βand interleukin-4, and measured the content of blood cytokines IL-1β,IL-4 by enzyme-linked immunosorbent assay (ELISA).Results The rats stool character of experimental group had changed from the 16th to 22th day after infection; compared with control group, the tuition threshold of intestinal were evident decreased(P<0.05);stool germiculture were negative,histology detection became normal.Through the macroscopic and histological scores, there were no significant difference between two groups(P>0.05);apply immunohistochemistry to detect the expression of CD11c,CD80,CD86 on terminal ileum, there were no significant difference between two groups(P>0.05);whereas the expression on distant colon, the experimental group were increased significantly(P<0.05);compared with control group, the expression of interleukin-1βon terminal ileum and distant colon of experimental group were increased significantly(P<0.05), and to detect the expression of interleukin-4, there were no significant difference between two groups (P>0.05). The expression of peripheral blood cytokines IL-1βof experimental group rats tended to increase compared with control group, but the difference was not significant (P>0.05) ; whereas the expression of blood cytokines IL-4 was reduced significantly (P<0.05).Conclusion (1) Established the rat model of post-infective intestinal function disorder successfully by Shigella flexneri intragastric.(2) The expression of peripheral blood cytokines IL-1βof rats which had become intestinal function disorder tended to increase, but the expression of blood cytokines IL-4 was reduced significantly. This indicated peripheral blood of rats exist immunoregulation disorder. (3) The expression of interleukin-1βon terminal ileum and distant colon of experimental group were increased significantly, but the espression of interleukin-4 had no evident increase, indicated the intestinal tissue of rats exist slight inflammation. (4) The expression of CD11c,CD80,CD86 on distant colon of experimental group rats increased significantly, whereas the expression on terminal ileum had no evident change, it indicated that the expression of Dcs on distant colon tend to differentiation , develop and mature, which made the function of distant colon immune tolerance reduced, promoted the immune reaction happened,influenced the disorder between intestinal anti-inflammatory and proinflammatory cytokines, made the intestinal function disorder.
