Objective: To Confirm that the synergistic inhibitory effects combination of Rapamycin with Geldanamycin in gastric cancer cell SGC-7901,which has a stronger inhibitation compared with rapamycin alone.Methods:The human gastric carcinoma SGC-7901 cell line was incubated with different drug groups for different time,MTT assay for cell growth and viability ,the cell apoptosis was measured by flow cytometry using annexin V/PI;immunocytochemistory was taken to test the espression of P-Akt.Results: Methylthiazole tetrazolium analysis showed that synergistic inhibition of combination of Rapamycin with Geldanamycin,the proliferation of cells was depressed in time and concentration–dependent manner;the expression of P-Akt was improved decreased as compared with either substance alone showed in the immunocytochemistory.the cell apoptosis percentage was increased obviously detected by flow cytometry.Conclusions: Geldanamycin improved growth-inhibitory and apoptosis-induction effects of the mTOR inhibitor rapamycin in gastric cancer cell SGC-7901 by inhibiting the activation of PI3K pathway that inhibited the activation of Akt;therefor combination of the two drugs had a synergistic effect in gastric cancer cells.
