Objective To detect the expression of Skp2 and p27kip1 in hypopharyngeal squamous cell carcinoma , then research their association with clinicopathological characteristics and the development and progression in Hypopharyngeal Squamous Cell Carcinoma. Methods The expression of Skp2 and p27 in 42 specimens of Hypopharyngeal Squamous Cell Carcinoma and 15 specimens of normal hypopharyngeal tissues were evaluated bv immunohistochemistry PV-9000 two-step methods..Results ( 1 ) The overexpression rate of Skp2 was significantly higher in hypopharyngeal squamous cell carcinoma (61.90%) than in normal hypopharyngeal tissues (26.67%);Skp2 overexpression was positively associated with T stage(r=0.329,P < 0. 05) and cervical lymph node metastasis (r=0.402,P < 0. 05) ;( 2 ) The overexpression rate of p27kip1 was significantly lower in hypopharyngeal squamous cell carcinoma (11.90%) than in normal hypopharyngeal tissues (53.33%) ; p27kip1 overexpression was negatively associated with T stage(r=-0.351,P<0.05) and cervical lymph node metastasis (r=-0.371,P<0.05);(3)The expression of Skp2 was negatively associated with p27kip1 in hypopharyngeal squamous cell carcinoma(r=-0.317,P<0.05)。Conclusion There is abnormal expression of the high expression of Skp2 protein and the low expression of p27kip1 protein in Hypopharyngeal squamous cell carcinoma. Skp2 expression might play an important role in the development and progression of hypopharyngeal squamous cell carcinoma to p27 proteolysis via ubiquitin proteasome pathway; To combined detect the expression of Skp2 and p27kip1 may be a great significance in the comprehensive assessment of the biological characteristics and estimating prognosis of patients with hypopharyngeal squamous cell carcinoma.
