Objective To investigate the expression and its clinicopathological significance of Cyclin D1,CDK4 and p57KIP2 in human laryngeal carcinoma.Methods The expression of Cyclin D1,CDK4 and p57KIP2 proteins were examined by PV-9000 2-step immunohistochemical technique in tumor tissues of 50 patients with laryngeal carcinoma. 15 cases polyp of vocal cord were used as control and comparison. Results The Cyclin D1,CDK4 protein positive rate in human laryngeal carcinoma were 60% and 54% respectively, which was higher than that in polyp of vocal cord tissue (P<0.05). Cyclin D1,CDK4 positive-expression were significant difference with clinical category,tumor differentiation and lymph node metastasis (P<0.05), but were not significant difference with different age,sex and clinical staging in laryngeal carcinoma patient (P>0.05). The p57KIP2 protein positive rate in human laryngeal carcinoma was 46%, which was lower than that in polyp of vocal cord tissue (P<0.05). The p57KIP2 protein positive-expression was significant difference with clinical category,tumor differentiation and lymph node metastasis (P<0.05), but was not significant difference with different age,sex,smoking history and clinical staging in laryngeal carcinoma patient (P>0.05). The expression of p57KIP2 was significant negative correlation with the expression of Cyclin D1,CDK4 . The expression of Cyclin D1 was significant postive correlation with the expression of CDK4.Conclusion Cyclin D1,CDK,p57KIP2 were close related in cell cycle. Over-expression of the Cyclin D1 and CDK4 protein and/or under- expression of the p57KIP2 protein may play an important role in the genesis , clinical category, differentiation, lymph node metastasis and progression of human laryngeal carcinoma. They shoud be used as important biological markers of malignant degree and lymphnode metastasis of laryngeal carcinoma. And the detection of Cyclin D1,CDK,p57KIP2 can be expected to used as prognosis prediction and new target of adjunctive therapy.
