| AIM:Establish femoral artery thrombosis model of rabbit with the adrenaline directly damage the vascular endothelial cells, to study the intervention function of laminaran L01 on the vascular endothelial cells derectly damaged by adrenaline, discuss the protect effect of laminaran L01 on the vascular endothelial cells through the morphological structure and anticoagulant function.METHODS:Rabbits were assigned randomly into five groups:saline group, adrenalin group, low dose and high dose laminaran L01 groups and enoxaparin group. The mixed liquor of saline, Adrenalin-saline, low dose and high dose laminaran L01-adrenalin and enoxaparin-adrenalin were injected into the dissociated femoral artery through an arterial cannula. Keep the liquor in the femoral artery for 25 minutes. Then, the arterial cannula was connected with a biological message recorder to show arterial pressure. The change of pressure curve to baseline presented the formation of thrombotic occlusion, which measured change of the anticoagulant function. Cut a segment of the femoral artery contacted with the drug and use the paraformaldehyde to fix, then made the tissue slicing and HE staining, observe the integrality degree of the vascular endothelial layer to measure the damage degree of vascular endothelial cell. Expression of TF in vascular endothelial was detected with immunohistochemical method,use the length ratio of undeciduous with the total vascular endothelial express the integrality of the vascular endothelial layer. Use and average gray value of the endothelial measure the express of the tissue factor on the endothelial and under endothelial, which can assess the damage degree in the other ways.RESULTS:The occlusion time of adrenalin group (19.99±1.92min) is significantly shorter than other groups.there are no statistical significances on occlusion time between saline group (57.38±3.85min) and enoxaparin group (54.69±4.35min) (P>0.05).The occlusion time in the low dose groups (33.04±3.05min) are shorter than high dose groups(46.94±4.31min)(P<0.01).The morphologic observation of femoral artery endothelial display that the endothelium are intact and smooth in saline, endothelium ruptures and breaks off in adrenaline group, and the endothelial are partly amotic in low dose laminaral LOl group, the endothelium in high and low dose of laminaral LOl groups are integrated and can see separation from the hypodermis, While coarse surface appears but not breaks off in enoxaparin group.Immunohistochemisty of the expression of TF in vascular endothelial layer and subendothelial tissue display that in adrenalin groups they are dyed in brown and manifest obscission are appeared in vascular endothelio; The dyeing of the vascular endothelial layer in saline group are buff and there are hardly tinctorial areas in this group;The enoxaparin group are dyed more darker than the saline group and there are a little tinctorial areas in the subendothelial tissue; The vascular endothelial layer of the low dose laminaran L01 group are dyed in brown madder and there are also some tinctorial areas in brown madder in subendothelial tissue;The vascular endothelial layer of the high dose laminaran L01 group are dyed in buff and the tinctorial areas are less in subendothelial tissue. The average grey value in adrenalin group (41.12±10.12) are lower than other groups (P<0.01),and there are no statistics significance between low dose laminaran L01 and high dose laminaran L01 groups (P>0.05).The integrated degree of vascular endothelial layer measured by pathology analyzer displayed that the integrated degree in high (64.35±3.39%) and low dose laminaran L01 (51.77±2.39%) groups are higher than the adrenalin group (38.13±3.40%) (P<0.01)CONCLUSIONS:The laminaran L01 can protect the vascular endothelial cells directly damaged by adrenalin and make the vascular endothelial layer integrality, decrease the expression of the tissue factor, play a role on the anticoagulant function through the protection of structure and function. |
