The traditional Chinese medicine -"Ye Dizhu"is the the root of Securinega suff ruticosa(Pall.) Rehd,which is a wide range of medicinal plant and is widely used for treatment of rheumatism, quadriplegia, back pain, limb numbness, paralysis, impotence, facial paralysis and diseases such as Children of malnutritioncodein.The modern pharmacological studies have indicated that the Securinega suff ruticosa(Pall.) Rehd has urges the central nervous system, specially the marrow and obvious exciting function. It to be able to elevate the blood pressure, cause the heart shrinks strengthens and the breath exciting. In addition, Securinega suff ruticosa(Pall.) Rehd also has the role of anti-tumor effect.So far, there are many researches in chemical constituents and pharmacological function of Securinega suff ruticosa(Pall.) Rehd. In order to make better use of this herb medicine, this experiment describes the extraction, isolation and Structure identification. Moreover, the determination method has been established for content analysis of virosecurinine in leaves of Securinega suff ruticosa(Pall.) Rehd in this paper. A series of tests proved that the determination method is accurate, reliable and reproducible to determine virosecurinine in the leaves of Securinega suff ruticosa(Pall.) Rehd.Extraction and SeparationDried leaves (3kg) were extracted by heat reflux in 95% Ethanol. The solvent volume were 20 L ,15 L and 10 L respectively for three times and the extraction time were 2h,1.5 h and 1h respectively. The ethanol was evaporated in vacuum and concentrated to cream. The cream was successively extracted by 1% of hydrochloric acid ,filtrated ,then the solution was neutralized by 5% of the sodium hydroxide solution, which was extracted by the organic solvent ethyl acetate, butanol respectively to obtain the extracts 32g and 82g in turn. The extracts was subjected to silica gel, polyamide, column chromatography separately using ethyl acetate–petroleum ether (1:1) , ethyl acetate– methanol (5:1) as mobile phase to afford six compounds, they are compound A (100mg),compound B (650mg),compound C (45mg),compound D (78mg),compound E and F.All the compounds were identified respectively by UV, IR, MS and NMR as virosecurinine (A),quercetin-3-O-α-L-rhamnopyranosyl-(1→6)-O-β-D- glucopyranoside (B),Octacosane(C),Dibutyl phthalate) (D),daucosterol(E) andβ-sitosterol(F), Compound B,C and D have been isolated from this plant for the first time Among them..Determination of virosecurinine in Ye Dizhu by HPLCIn this paper, we present a more sensitive and accurate quantification method by HPLC for determining the major alkaloid (compound A-virosecurinine) in the Securinega suff ruticosa(Pall.)
