Object:To study expression of MMP-9 and TIMP-1 in placenta trophoblasts of severe preeclampsia, and to discuss their function in pathogenesis and development of pregnancy induced hypertension, as well as their influence on fetal weight.Method:Choose and collect 20 pieces of placenta from both normal pregnant women and those with severe preeclampsia, respectively, which were obtained in caesarean delivery in the 1st,2nd and 3rd Hospital of Jilin University. Use two-step immunohistochemistry method to dye the samples in order to detect expression of MMP-9 and TIMP-1.Result:the mean grey scale of immunohistochemistry dying of MMP-9 in the group of severe preeclampsia is higher than and significantly different from the group of normal pregnancy(P<0.05), and that of TIMP-1 is higher in normal pregnancy group, but not significantly different, and the ratio of MMP-9/TIMP-1 is lower in normal pregnancy group, but not significantly different between the two groups, either. The expression level of MMP-9 and TIMP-1 is negatively related, but the relationship is not significant, due to the small number of samples collected, perhaps. Incidence of fetal growth restriction is significantly different between the two groups. The ratio of MMP-9/TIMP-1 and new born weight/mean new born weight of the same fetal age has a significantly positive relationship.Conclusion:1) Expression of MMP-9 and TIMP-1 is mainly concentrated in cytoplasm and cell membrane.2) Expression scale of MMP-9 in placenta trophoblasts is significantly different between normal pregnancy and severe preeclampsia groups, which is lower in the later group.3) Expression scale of TIMP-1 in placenta trophoblasts is not significantly different between the two groups, but the mean scale of the normal pregnancy group is lower.4) The decrease of MMP-9/TIMP-1 ratio may play an important role in pathogenesis and development of pregnancy induced hypertension. The expression level of MMP-9 and TIMP-1 is negatively related. 5) Incidence of fetal growth restriction is higher in severe preeclampsia group, and there is a close relationship between the ratio of MMP-9/TIMP-1 and fetal growth.
