[Objective]Periodontitis is a bacterial infectious disease. Periodontal pathogen destroys periodontal tissue directly or indirectly. Quantity of bacteria in the inflammatory site may be a key factor in the development of Periodontitis. This study quantifies the main periodontal pathogen in the saliva, supragingival and subgingival plaque samples from patients with periodontal diseases to investigate the distribution of bacteria in the different site and the relationship between the quantity of periodontal pathogen and periodontal status. Change of periodontal status before and after mouthrinse usage is also observed for providing cues in the daily prevention of periodontal diseases.[Methods]50 patients with periodontal disease and 12 periodontal healthy person without systemic diseases were included. Disposable paper cup. probe and 30# standard paper point were used respectively to collect saliva, supragingival and subgingival plaque samples before and after using mouthrinse for 7 days. Pocket depth (PD), clinical attachment level(CAL), bleeding on probing (BOP) and BOP after mouthrinse usage were recorded. Observe the change of periodontal status and quantify the main periodontal pathogen acfinobacillus actionmycetemcomitans(Aa). porphyromonas gingivalis(Pg). fusobacteriurn nucleatum(Fn), prevotella intermedia(Pi) in the samples before mouthrinse usage with real-time PCR.[Result]In patients with periodontal diseases, the sites of BOP were reduced significantly after using mouthrinse for 7 days(P<0.05). Compared with control group, quantity of main periodontal pathogen in periodontal disease patients is rise except Fn in the saliva and supragingival plaque. Pg in saliva samples has highly significant differences(P<0.01). Pi,Pg in supragingival plaque and Fn,Pi,Pg in subgingival plaque has significant differences (P<0.05). main periodontal pathogen in the saliva and subgingival plaque was higher when PD≥3mm. Quantity of Aa,Fn in the saliva and Fn in subgingival sample rises significantly (P<0.05)[Conclusion]In patients with periodontal diseases, quantity of main periodontal pathogen Aa,Fn,Pg. Pi in subgingival plaque and all other main periodontal pathogen except Fn in saliva and supragingival plaque was higher than the control group. Aa,Fn in saliva and Fn in subgingival plaque increase significantly as the PD increases. Using mouthrinse could reduce bleeding symptom without changing the oral hygiene habit.
