| ObjectiveTo investigate the effects of mesenchymal stem cells(MSCs) transplanting on recovery of neurological functions in cerebral ischemia-reperfusion in rats and its molecular mechanism. by observing the changes of surviving,caspase-3 protein, study the relationship MSCs transplanting and inhibit cell apoptosis and promote functional recovery in cerebral ischemia-reperfusion in rats.MethodsA total of 96 healthy adult male SD rats were randomly divided into normal group(n=6), sham operation group(n=6), PBS group(n=36)and MSCs treatment group(n=36), and rats of the last two groups were established cerebral ischemia-reperfusion model with suture emboli method, PBS group and MSCs group, with separately injection of PBS and MSCs 3 h after MCAO via Stereotaxic Instrument. At 1 d,3 d,6 d after reperfusion, the neurological deficits of rats in every group were evaluated, and cerebral ischemia tissue was obtained to detect the cell apoptosis by TUNEL method, and the expression of survivin and caspase-3 proteins, using immuno-chemistry and western blot.Results3 d after MCAO, Brain infarction area was differentiated by TTC dyeing. The result showed that control group of imitational operation had no infarction areas.The volume of cerebral infarction in MSCs group reduced significantly compared with PBS group,(all P< 0.01);The neural cell apoptosis, the expressions of survivin and caspase-3 proteins of sham operation group were negative. Compared with PBS group, the neurological scores of MSCs treatment group significantly decreased, the number of apoptotic neural cells and caspase-3 were lower, while the expression of survivin significantly increased (P<0.05).ConclusionMSCs treatment could improve the neurological function of cerebral ischemi-reperfusion in rats.MSCs treatment could up-regulatin survivin protein expression and down-regulating caspase-3 protein expression.MSCs treatment could reduce apoptosis of neural cells. |
