OBJECTIVE:By detecting the effect to the number of bone marrow stem cell cycle,bone marrow stem cell cycle and the level of TPO mRNA and c-Mpl mRNA expression from marrow suppression mice in the ways of tonifying kidney and tonifying spleen, to study the protective therapeutic action and mechanism of Liuwei Dihuang Decoction,Buzhong Yiqi Decoction and Compound Danshen Decoction on the marrow-suppressed mice. And then provide the basis for clinical medicine.METHODS:100 mice were randomly divided into 5 groups, namely normal group, control group, Liuwei Dihuang Decoction group,Buzhong Yiqi Decoction group and Compound Danshen Decoction group, with 20 rats in each group. Between human and mice by dose converted to 0.2mL·10g-1 of the dose administered to experimental mice in each group, model control group fed with distilled water. The mice were intraperitoneal injected after routine disinfection with 100mg/kg cyclophosphamide in the 9th,11th,13th days during the experiment. Flow Cytometry was adopted to investigate bone marrow cell cycle and the mount of the bone marrow, the expression of the TPO mRNA and c-Mpl mRNA was detected by RT-PCR technique simultaneously.RESULTS:1. On the 14th day of the experiment, compared with normal control group, the mount of the bone marrow of model control group was increased with statistically significant (P<.01); Compared with model control group, the mount of the bone marrow was raised with statistically significant in the Buzhong Yiqi Decoction group and Liuwei Dihuang Decoction group. On the 28th day of the experiment, compared with normal control group, the mount of the bone marrow of model control group was increased with statistically significant (P<0.01); Compared with normal control group, the mount of the bone marrow was raised with statistically significant in the Buzhong Yiqi Decoction group and Liuwei Dihuang Decoction group.2. On the 14th day of the experiment, compared with normal control group, the proportion of G1 of the bone marrow cell cycle was raised with statistically significant (P<0.05) in model control group, the proportion of S and G2 of the bone marrow cell cycle was increased with statistically significant (P<0.01) in model control group; Compared with model control group, the mount of the bone marrow was raised with statistically significant in the Buzhong Yiqi Decoction group and Liuwei Dihuang Decoction group. On the 28th day of the experiment, compared with normal control group, the mount of the bone marrow of model control group was increased with statistically significant (P<0.01); Compared with normal control group, the mount of the bone marrow was raised with statistically significant in the Buzhong Yiqi Decoction group and Liuwei Dihuang Decoction group.3. On the 14th day of the experiment, compared with normal control group, the expression level of TPO mRNA in spleen of mice, through which they can accelerate the marrow cells of marrow-suppressed mice was increased with statistically significant (P<0.01) in model control group, the expression level of c-Mpl mRNA in spleen of mice was increased with in model control group, but not statistically significant; Compared with normal control group, the expression level of TPO mRNA and c-Mpl mRNA were raised with statistically significant. On the 28th day of the experiment, compared with normal control group, the expression level of TPO mRNA was increased with statistically significant (P<0.01) in model control group, the expression level of c-Mpl mRNA was increased with in model control group, but not statistically significant; Compared with normal control group, the expression level of TPO mRNA and c-Mpl mRNA were raised with statistically significant (P<0.05).CONCLUSION:1. Buzhong Yiqi Decoction and Liuwei Dihuang Decoction may increase bone marrow nucleate cells of anemia mice.2. Buzhong Yiqi Decoction and Liuwei Dihuang Decoction promote bone marrow nucleate cells entering proliferative phase and stimulate proliferation of cells.3. Buzhong Yiqi Decoction and Liuwei Dihuang Decoction can promote gene expression of TPO mRNA and c-Mpl mRNA in anemic mice. It maintain survival and induce differentiation of hematopoietic progenitor cells through interference to cytikin/recepter single pathway on gene level.
