ObjectiveTo investigate the therapeutic effect of Qingyan mixture on patients with laryngeal cough of lung-dryness due to deficiency of YIN type by clinical observation, and explore the mechanism of Qingyan mixture in sore throat relieving, inflammation eliminating and cough inhibition by animal experiments.MethodsSixty patients with laryngeal cough of lung-dryness due to deficiency of YIN type were divided into treatment group and control group,30 cases per group from January 2009 to January 2010 in respiratory outpatient department. Patients in treatment group took Qingyan mixture 30ml every time and three times a day, while patients in control group were treated with Ketotifen lmg bid, carbetapentane 25mg tid. The course of treatment was 10 days. Therapeutic effect was evaluated by the changes in clinic symptoms and objective signs. The animal experiments of Qingyan mixture were carried out by 15% ammonia modeling SD model of acute pharyngitis, the model of mouse ear edema inflammation was induced by apply of xylene, and the model of acute cough was built up by spraying ammonia in mice.ResultsPartⅠ:Clinical Research1. therapeutic effects:In treatment group,6 patients were cured,14 patients had markedly improvement, 8 patients partly cured and 2 patients failed in treatment. The number of the cured, markedly improvement, partly cured and treatment failed in control group was 1,7,17 and 5 respectively. The total efficiency of treatment group was 93.3%, which was significant higher than control group (83.3%), P<0.01.2. the comparison of the single clinical symptoms or objective signs: The comparison of single clinical symptoms or objective signs before and after treatment:In the dry cough, the treatment group had better effects than the control one (P<0.05); there were no significant differences in throat, pharyngeal itching, foreign body sensation in the throat, and pharyngeal signs beween two groups (P> 0.05).PartⅡ:Animal Experimental:1. Acute pharyngitis model in SD rats:Qingyan relieved the inflammatory reaction of pharynx in SD rats with acute pharyngitis induced by 15% ammonia spray. The pathological changes were more significant in medium and high dose groups, which were similar to the results of positive control group deal with Guilin watermelon frost.2. Xylene-induced ear edema inflammation model in KM mice: Qingyan mixture significantly inhibited the ear-edema induced by xylene. The inhibiting effect was more obvious in high and medium dose group, which were similar to the results of positive control group.3. Strong aqua induced cough model in KM mice:Strong aqua was used to evoke the cough reflex of KM mice. After 5 days of intragastric administration with different dose of Qingyan mixture, all mice had shorter cough latent period and less cough frequency, which were significant differences beween the control group(intragastric administration with normal saline) and positive control group(intragastric administration with carbetapentane citrate).ConclusionsQingyan mixture can relieve the clinical symptoms and objective signs in patients with laryngeal cough of lung-dryness due to deficiency of YIN type. Animal experiments demonstrate that Qingyan mixture can relieved the inflammatory reaction and cough reflex. Qingyan mixture can used in the treatment of patients with laryngeal cough of yin-dryness type.
