I. lophanthoides var. gerardianus (Bentham)H. Hara,I. lop-hanthoides var. graciliftorus (Bentham)H. Hara andI. serra (Maxim.)Kudo are medicine as R. serra in folk.Ⅰ. lophanthoides var. gerardianus (Bentham)H. Hara in this paper refer to the dried aboveground of I.lophanthoides var. gerardianus (Bentham)H. Hara from labiatae.Many pilot studies prove that Water-soluble part of I. lophanthoides var. gerardianus (Bentham)H. Hara is the active ingredients. So this paper mainly research on extraction and purified technology of Water-soluble flavonoids.Literature Research:In this paper, biological characteristics, germplasm resources, biological form, identification, chemical constituents, pharmacological activity studying of I. lophanthoides var. gerardianus (Bentham)H. Hara are summarized,also the research situation of flavonoids, including the extraction and purification methods, pharmacological activity research, in order to further design and implementation of the experimental scheme provides theory basis.Systematic pre-experiment:the components in I. lophanthoides var. gerardianus (Bentham)H. Hara contain amino acid, terpenes, flavonoids and phenols etc. which based on systematic pre-experiment, chemical identification and the extracts by water. Flavonoids and anthraquinone are mostly among these components. Combined with the result of research on pharmacology and pharmacodynamics of I. lophanthoides var. gerardianus (Bentham)H. Hara, the research content of this paper contains mainly aim at Water-soluble flavonoids compounds.Separation of components:Filled chromatography column respectively by macroporous resin and extracting from water-saturated butanol, and the latter is used for isolate monomet components, chloroform:methanol (100:0-0:100) is used as eluent, in order to guide the utilization of eluent, use thin-layer chromatography to trace the spot. Incorporate samples that contains the same spot, concentrate them for crystallization, then use TLC or HPLC to detect it, if purity is low or having more than one spot in the samples, use the preparative liquid to separate this two spots, finally, according to physical-chemical reaction and analysis the data of spectrums (ultraviolet, infrared, mass spectra, nuclear magnetic resonance), the structures of the component was identified, that it is 6-C-arabinssyl-8-C-glucosyl apigenin.Extraction technology of Water-soluble flavonoids:Single factor exploration determined single factor level, the threee factors are as follows: extraction time, amount of solvent, extraction times. Select factor and level for orthogonal design L9 (34), study the three factors:extraction time, amount of solvent and extraction times, and how they effect the extraction efficiency. Finally determine the optimum extraction factor of I. lophanthoides var. gerardianus (Bentham)H. Hara:twelve times water, extract three times, each 2.0 hours.Purified technology of Water-soluble flavonoids:select five macroporous resins that have different polarities (their types are:FL-1,DN-603,HPD-826,HPD-722,HPD-600), study the static and dynamic adsorption properties of macroporous resin, also research on how these factors affect the enrichment of total flavonoids, the factors are the concentration of liquid filled column, flow velocity of upper samples, divulging curves, elution solvent and elution amount, according to which the optimal technology for enrichment is determined and then to verify it.The optimal technology parameter of macroporous resin type HPD-600 is as follows:the concentration of total flavonoids flow through the column is 2.27- 2.52mg/mL, flow velocity of upper samples is 2BV/h, the volume of upper samples is no more than 6BV, the water amount used for wash impurities is 4BV, elution solvent is 60%ethanol, elution amount is 5BV. Eluate from 60%ethanol thorough macroporous resin type HPD-600 is tested, the purity of total solids in total flavonoids is as high as70.21%, refining degree is 1715.96 %.
