Populus canadensis Moench. is a genus of Sect. Aigeitos Duby plants. Populus canadensisMoench. was introduced into China in the mid-19th century. Because of its fast growth,easy reproduction,and adaptability, Populus canadensis Moench. was widely planted in China.In this paper,carotenoids and flavonoids in Populus canadensis Moench. leaves were studied.Firstly, the extracting and determing methods of carotenoids were introduced. The single factor test and the orthogonal experimental design were carried out to optimize the extraction technology of total carotenoids from leaves of Populus canadensis Moench..The result indicated that using petroleum ether as extraction solvent,the optimal extraction technology of total carotenoids was as follows:500W of ultrasonic power and 40:1 of the ratio of solvent to material, temperature of 55℃,30min of extracting time. Under the optimum conditions, total carotenoids content was 50.57mg/100g.Secondly, the method for determination of rutin in Populus Canadensis Moench. leaves was developed by High Performance Capillary Electrophoresis(HPCE).Sample was extracted with methanol by ultrasonic.The optimum determination conditions were investigated experimentally and the results were as follows:the running buffer of 50 mmol/L borate(pH=9.3),injection time 5s with pressure,tempe-rature of 25℃,applied voltage of 20 kV,detection wavelength of 254 nm.The developed method was utilized to investigate variation of the content of rutin during the growth of Populus Canadensis Moench. Leaves,and the content of rutin in October is the most.Thirdly, the relationship between the content of flavonoids of Populus Canadensis Moench. leaves and other relevant components was investigated.
