Ziziphus jujuba Mill.,belonging to the Ziziphus genus,Rhamnaceae family,and is native to our country.Fruit and leaves of Z.jujuba for this study were collected from Suide,Yulin,Shaanxi Province,China.A valid HPLC-DAD method was established to quantitatively analyze sixteen compounds in the leaves of Z.jujuba,which was comprised five ripening stages from six cultivars in this region.It was the first time to study the seasonal variability and the differences among cultivars of chemical constituents in Z.jujuba leaves.Based on the aforementioned work,it was the first time to study the hepatoprotective activity of Z.jujuba leaves's extract.In addition,anticancer activity of 18 compounds against four type tumor cell lines(MCF-7,A549,HepG2 and HT-29)was conducted.The results demonstrate that:(1)The major ingredients of Z.jujuba leaves were flavonoids with the total contents of the nine flavonoids in 30 samples ranged from 18.52 mg/g to 39.51 mg/g.The content variation trends and differences between cultivars of constituents in Z.jujuba leaves have been achieved,and June is the best time for the sampling of Z.jujuba leaves,which can be used as the resource of flavonoids.HPLC fingerprint file of Z.jujuba leaves was set up for the first time.Furthermore,30 samples have divided into two main clusters by hierarchical cluster analysis.(2)EtOH-water extract of Z.jujuba leaves exhibited strong hepatoprotective activity to the hepatic damage in mice,which induced by CCl4.EtOH-water extract of Z.jujuba leaves can lower liver index,serum ALT and AST,decrease hepatic MDA,and enhance SOD and GSH-Px level.Hepatoprotective effects of EtOH-water extract were also confirmed by histopathological observations,and flavonoids might act as the active ingredients for biological activities of hepatoprotection.(3)Compounds 3-O-(traps-p-coumaroy)-alphitolic acid(1),3-0-(cis/p-coumaroyl)-alphitolic acid(2),3?-O-(trans-p-coumaroyl)-maslinic acid(3),benthamic acid(6),terminic acid(7),betulinic acid(9)and apigenin(12)had strong growth inhibitory effects and induced cell death on the four cancer cell lines,and six among them is triterpenoids.
