| In recent years, N-linked oligosaccharide is correlated with cell cancer, metastasis and hematopoietic function. Therefore, the relationship between structure and biological function has been recognized by many reseachers. At the same time, it is extremely necessary to develop analysis instruments and seek a new analysis method, which is simple, sensitive and higher selective.Endo-N-acetylglucosaminidase (Endo-M) isolated from Mucor hiemalis, has a feature, which can severed the GlcNAc-GlcNAc bond in oligosaccharides. The result is that one GlcNAc retain on the glycopeptides, and the other released is combined with acceptor to become a new compound. For the characteristic of Endo-M, we designed and synthesized a kind of acceptor for detection of N-linked oligosaccharides, and these acceptors include GlcNAc and ionic. We found the optimization method for synthesis and purification of these acceptors. Furthermore, we also researched these acceptors for the resolution of N-linked oligosaccharides in glycopeptides, based on enzyme transglycosylation reaction. They can improve the sensitive for detection N-linked oligosaccharides by MS because they are either easily ionized or have charge. We also studid the real sample and use pronase E distill N-linked oligosaccharides from ovalbuminl And we also researched how many kinds N-linked oligosaccharides in ovalbumin.A new derivative reagent PDPZ synthesized is used to examine carboxylic acids. Simultaneously, a best derivation condition has been found and a simple and rapid HPLC detection method has been also devised. |
