Single cell gel electrophoresis (SCGE), or comet assay established by Ostling, modified by singh, is used to detect DNAdamages of karyocyte, which can detect DNA damages in eukaryotic individual cells. Some papers reported that compared to chrosome aberration and micronuclei test, concerning radiation workers'DNA damages estimation, the outcome of SCGE is homologous. Our research aimed at:â‘ To explore a new, simple and quick radiation biodosimeter, which can be applied to estimate biological dose in case of radiation accident, and acquire dose-effct curse;â‘¡Explored time effectiveness of DNA repair after irradiation, and described curves and plane of DNA repair, then improved the accuracy of estimation of radiation biodosimetry after radiation accident, and acquire repair curve;â‘¢To investigate the status of DNA damages of radiation workers, and discuss the relationship between the different types of work or work time and the level of DNA damage, and explore the feasibility of neutral single cell gel electrophoresis serving as a index of radiation workers'physical examination.â‘£To explore the feasibility of using alkaline single cell gel electrophoresis to rapid estimate the dose of radiation damage.With the development of Molecular biology of radiation, more and more research of irradiation came toward DAN damages and repairs. The detection method of DNA strand breaks, especially double stand breaks is a critical technique. Single cell gel electrophoresis (SCGE) established in the eighties is a sensitive method usd to detect DNA damages and repairs.The contents of our research:â‘ DNA double-strand break were detected using neutral single cell gel electrophoresis(SCGE), and all the indexes of comet assay including TDNA%, TM and OTM were analyzed by CASP(Comet Assay Software Project). The curve of dose-efiect was fitted using SPSS 16.0 software;â‘¡Detected DNA double-strand break and repair in lymphocytes of human using neutral single cell gel electrophoresis (SCGE) after different dose of radiation in 3h,24h,48h,72h and estimated curves and plane of DNA repair;â‘¢The objects were divided into control group and radiation group, and the latter was divided into doctors at radiation department group, cardiologists group and orthopaedic surgeons group according to types of work, and<10 group,10-group and 20-36 years ole group according to work time. The neutral single cell gel electrophoresis (SCGE) was used to detect the DNA damage of control group and each radiation group.â‘£using alkline single cell gel electroporesis to detect DNA damage of peripheral lymphocyte, irradiated by the of human peripheral blood. The test outcome indicatedâ‘ Statistically significant dose-efect relationships were observed in all the indexes of comet assay, OTM was superior to other index.â‘¡The curves of DNA repair in vitro of human lymphocytes presented nice model and the repair of radiation-plane presented good smoothness.â‘¢The parameters of DNA damage in radiation group are significant different from control group, for example, the TDNA%, TM, OTM were much higher than those of control group. Significant difference was found among different types of work or different work time, and the invasive cardiologists group is higher than doctors at radiation department group and the latter higher than orthopaedic surgeons group.â‘£the outcome of test in vitro indicated that there was remarkable difference beteen the control group and the radiation group(p< 0.05). The tail moment(TM) value increased with the dose of radiation and there was good linearity of dose-effect. Y=0.3619+2.1834D(r=0.9946).Based on the outcomes, it is the conclusion thatâ‘ Application of neutral comet assay combined with CASP analysis could serve as a new radiation biodosimeter;â‘¡Curves of repair under the different doses showed a linear relationship, repair of radiation-plane model can be used as reference of primary injury of DNA double-strand break.â‘¢Long-term and low dose radiation exposure can result in DNA damages of radiation workers increasing. Obvious relationship was found among different work time groups.â‘£The above outcomes show that alkline single cell gel electrophoresis is a effective method for detecting DNA damage, and have good dose-effect relation, but DNA damage repair is so fast that the range of time is limited.The innovation in this study is the use of the neutral Single cell gel electrophoresis, which mainly detect DNA double-strand break, and the latter is more suitable for the detective subject because it is more stable than the single-strand break.
