Backgroud:Tumor metastasis is a significant contributor to death in cancer patients. Despite recent improvements in the treatment of cancer, metastatic recurrence remains a tremendous clinical hurdle.Metastases arise when cancer cells leave the primary tumor site and form new tumors elsewhere. The hypothesis that metastasis might also involve loss of gene functions that maintain the normal state of a cell emerged after the identification of RB as a tumor suppressor gene. Metastasis suppressor genes (MSGs) are defined by their ability to inhibit overt metastasis in a secondary organ without affecting tumor growth at the primary site. Over 20 MSGs have been confirmed. The expression of KISS1 gene may play important role in inhibiting metastasis of breast cancer,Loss of KiSS1 gene expression that leads to the invasion and metastasis of breast cancer may participate in the lymph node metastasis. KAI1 may play an important role in cancer metastasis.KAI1 is a prognostic indicator of disease-free survival in breast cancer patients and demonstrates the ability to play a role in governing the metastatic nature of breast cancer cells.FHL1 may play an important role in ER signaling as well as breast cancer cell growth regulationObjective:To investigate relationship between expression of metastasis suppressor genes KISS1,KAI1and FHL1 and lymph node metastasis in breast cancer.Methods:Tissue microarray and immunohistochemistry by EnVision methods were employeed to study 59 archival breast carcinoma samples and 31 noncancerous tissue samples. The presences of KISS1,KAI1,and FHL1 protein were investigated in relation to lymph node metastasis.Results:(1) The expressions of KISS1,KAI1 and FHL1 were insignificantly different between breast cancer tissue group and noncancerous tissue group(P>0.05);(2) The expressions of KISS1,KAI1 and FHL1 were negatively related to axillary lymph node metastasis and clinical stage(P<0.05), but were not statistically correlated with age,tumor size in breast carcinoma tissues(P>0.05); the expressions of KISS1 and KAI1 were positively related to P53(P<0.05)the expression of KAI1 was positively related to ER and PR(P<0.05); the expressions of FHL1 were negatively related to PR(P<0.05).(3) The expressions of KISS1,KAI1 and FHL1 were positively related to each other(P<0.01).Conclusions:The lowexpression of KISS1,KAI1 and FHL1 are closely correlated with the tumor invasion and metastasis of breast carcinoma. Detecting these indexes can be helpful in assessment of the invasional and biological behaviors of breast carcinoma.
