Objective:To research the inhibition effects of lobelia chinensis lour decoction (LCLD) in H22 tumor-bearing mice, influence to the expression of P53% C-erbB-2 and PCNA, and explore the possible anti-tumor mechanism.Methods:After injected H22 hepatoma cell line into their right oxter, mice were randomly and equally divided into three groups:normal saline (NS),lobelia chinensis lour decoction (Chinese traditional medicine) and 5-Fu(Western medicine) groups. Mice of NS group were given normal saline, mice of Chinese traditional medicine were given LCLD,and those of Western medicine group were injected with 5-Fu.All animals were fed for 14 days continuously. The 15th day mice were killed by cervical vertebra dislocation and tumor were taken out and weighted.Then the anti-tumor rate was calculated; Slices were made for immunohistochemical analysis on P53,C-erbB-2 and PCNA expression.Results:1.The inhibition effects of LCLD on primary hepatic carcinoma showed:Tumor weight of control group (n=8) was 0.91±0.51g, while the tumor weight of LCLD group (n=8) was 0.60±0.11g, the inhibition rate was 33.98%, it was obviously lower than control group,meanwhie the differences between LCLD group and control group were statistical significance (P<0.05). Tumor weight of 5-Fu group (n=8) was 0.45±0.11g, the inhibition rate was 50.48%, it was obviously lower than control group the differences between 5-Fu group and control group were statistical significance (P<0.01).2.Expression of P53 in H22 cells of three groups by immunohistochemistry staining had positive expression. One-way ANOVA and LSD test showed that the expression of P53 in LCLD and 5-FU group were significantly increased and different from that of control group (P<0.05, P<0.01), while the differences between LCLD group and 5-FU group were no statistical significance (P>0.05).3. Expression of C-erbB-2 and PCNA in H22 cells of three groups by immunohistochemistry staining all had positive expression. One-way ANOVA and LSD test showed that the expression of C-erbB-2 and PCNA in LCLD and 5-FU groups were both significantly decreased and different from that of control group (P<0.05, P<0.01), while differences between LCLD and 5-FU were not statistically significant (P>0.05).Conclusion:1.LCLD group had apparent inhibition effects on H22 hepatoma cell line.2.LCLD up-regulated expression of P53,it promoted tumor cells apoptosis and prevented the growth of malignant cells by a mechanism that it inhibited P53 from regulating the growth of cells at the level of DNA replication.3.LCLD suppressed the expression of C-erbB-2;it inhibited cancer cells from dividing and synthesis of DNA and protein.4. LCLD down-regulated expression of PCNA; it inhibited hepatocarcinoma cells Proliferation by inhibiting their activity.
