Objective To investigate the roles of lobelia on proliferation and apoptosis of EGF-induced HaCaT cells,to observe the effect of lobelia on the expression of gene cyclin D1 and Bcl-2 in HaCaT cells and to explore the mechanism of lobelia in the pathogenesis of psoriasis.Methods MTT assay was used to detect the effect of different concentrations of EGF on the proliferation of HaCaT cells and different concentrations of lobelia on the proliferation of EGF-induced HaCaT cells;Cell apoptosis rate was evaluated by FITC Annexin and V/PI staining of flow cytometry;The expression of cyclin D1 and Bcl-2 protein were detected by western blot assay.Results(1)The proliferation of HaCaT cells was obviously promoted by 5 ng/ml of EGF(P<0.05),the effection of proliferation was nearly maximum when the concentration of EGF was 10 ng/ml(P<0.01);When the concentration of EGF was 20 ng/ml to 120 ng/ml,the ability of proliferation was no statistical significance than 10 ng/ml of EGF(P>0.05).(2)After cells were treated with lobelia(5 mg/ml-60 mg/ml),HaCaT cells growth were inhibited(P<0.01)in a dose-dependent manner.(3)Cell apoptosis can be induced in a dose-dependent manner on 10 mg/ml,20 mg/ml,30 mg/ml of the lobelia concentration,and when the concentration of lobelia was 20 mg/ml the effection of cells apoptosis was the best.(4)western blot detection showed that after HaCaT cells were treated with lobelia(5mg/ml,10mg/ml,30mg/ml)the expression of cyclin D1 and Bcl-2 was gradually decreased.Conclusion Lobelia inhibited the cells proliferation and induced cell apoptosis in EGF-induced HaCaT cells.lobelia may play the role of inhibiting proliferation and promoting poptosis on KC by reduceing the expression of pro-proliferation gene cyclin D1 and anti-apoptotic gene Bcl-2.
