| Objective:Hepatocellular Carcinoma is one of common cancer for human. There were nearly one million people have been died of the Hepatocellular Carcinoma every year in the word, and 45% of the amount were Chinese. Therefore, the treatment of the Hepatocellular Carcinoma is one of the hot point for the researchers. Nuclear factor-κB is a common transcribe factor in Eukaryotic, and it play a very important role in disease, such as inflammation, immunity, tumor and so on. The signal way of the NF-κB is very complex, and is the object of the experiment. Some researches reported that NF-κB have a high expression in many tumors, and it play a role in the tumors cause and the next steps cooperate with other cell factors. There are other investigations reported that huangqi have the function of inhibiting the activity of NF-κB. This experiment investigated the huanqi injection'function on the Hepatocellular Carcinoma Cells-HepG2 through detecting the expression of NF-κB protein, surveyed the cells'proliferation and apoptosis, investigated the content of NF-κB in HepG2 cells, and to give the theory of supporting in clinical treatment of Hepatocellular Carcinoma.Methods:We cultured the HepG2 cells and experiment in logarithm-grow period. Extractive of traditional Chinese medicine- huangqi injection effected on Hepatic cancer cells, and then used the MTT method to detect the cell's sensibility about the consistency ladder of Huangqi injection. The whole experiment was separated to six grops:marked A,B,C and D, group A was the control group, B,C,D,E,F were the test groups and the concentration was 6.25%,12.5%,25%,50%,75%, respectively. We detected the OD numerical value after 48 hours. We accounted IC50 according to the value of the OD. At last, we choose control groupA,B,C,D test groups (the concentration was 6.25%,12.5%,25%) as the investigating groups in experiment. We surveyed the exterior appearance of the HepG2 cells, extracted the NF-κB protein. NF-κB protein expression was detected according to the groups of consistency ladder of Huangqi injection by the method of Western blot. We analysis the result by software SPSS 11.5, the data was revealed by x±S, and the method is Dunnett test, P<0.05 is the test standard.Results:After 48 hours affect of huangqi injection on Hepatic cancer cells --HepG2,the result of MTT illustrated that:different groups have corresponding affects on inhibiting the Hepatic cancer cells'proliferation. More over, Hepatic cancer cells'proliferation was inhibited much more obviously according to the arising concentration ladders. The result was OD value of group A:1.2212±0.0746;compared with group A, group B OD value 1.1177±0.08597 (P>0.05), group C OD value 1.0853±0.06371 (P<0.05), group D OD value 0.7720±0.05742 (P<0.01).separately. IC50=0.39192. The content of the cell:cells in group A had the whole structure and was like polygon; cells in group A was swelled, the shap of polygon is disappeared; cells in group C was much more swelled than B, and the cell was scatterd; the amount of cells in group D was the least, the shape of them was irregular. the method of Western blot showed that:the protein of NF-κBp65 was declined by 8.0%,32.1%,46.9% according to the group A in group B,C,D, respectively. There was no discrepancy ofβ-actin between groups.Conclusion:Huangqi injection have the function to inhibit the proliferation of human Hepatic cancer cells --HepG2,and have the active function in changing the structure of the cell, the result was more obvious according to the higher concentration. Huangqi injection also can decline the NF-κB protein expression, and it was concentration dependent. |
