OBJECTIVE: Using Astragalus injection to treat tumor cells and observe its proliferation to select experimental cells.METHODS: Using CCK-8 method to detect the growth inhibition rate of tumor cells which was treated by Astragalus injection after 24 hours,48 hours and 72 hours.RESULTS: Calculating the growth inhibition rate of tumor cells which was treated by Astragalus injection?Lung cancer A549 cells' growth were promoted at low drug concentration,but inhibited at high drug concentration,and there is good consistency of time and concentration.For hepatoma HepG2 cells,hepatoma hep3 B cells,and neuroblastoma SH-SY5 Y cells,the growth was inhibited by treatment of Astragalus injection after more than 24 hours,and without growth-promoting effect.However,at 24 hours,most of the drug concentration groups showed significant cell growth promoting effects,which were inconsistent with the effects exhibited at other times.CONCLISION: Considering the good consistency of time and concentration of Astragalus injection,lung cancer A549 cells were selected as experimental subjects for this follow-up experiment.OBJECTIVE: To detect the biological behavior of A549 cells,which was treated by Astragalus injection.METHODS: Using microscope to observation the morphological changes in cells.Calculating the half maximal inhibitory concentration(IC50)by Modified Kou's method.Observing the ability to move in both horizontal and vertical directions by Wound-healing Assay and Transwell Migration Assay.The cell invasion assay was used to observe the changes of cell invasion ability.The changes of cell apoptosis were observed by Flow Cytometry(FCM).RESULTS: After treated by high concentration of Astragalus injection,the A549 cells were shrinkage and gradually detached from the adherence.The IC50 at 24 h,48h,and 72 h was calculated as: 776mg/ml,851mg/ml,616mg/ml.In Wound-healing Assay,the scratches markedly healed in the groups of control group,100 mg/ml and 200 mg/ml concentration groups.Because of the detachment of cells in the 400 mg/ml,600 mg/ml and 800 mg/ml concentration groups,we cannot accurately measure scratch width.In Transwell chamber migration and invasion experiments,the number of cell through the membrane in the 100 mg/ml concentration group was more than the control group.However,the number of cell through the membrane gradually decrease with the increase of concentration.In the apoptosis experiment,as the drug concentration increases,the number of apoptotic cells gradually increases.CONCLISION: Astragalus injection can promote the growth of A549 cells at low concentrations.With the increase of the concentration,the growth inhibitory effect gradually appears.
