Objective: To study the effects of microRNAlet7,Ras,HMGA2 and C-myc on carcinogenesis and progression in non-small cell lung carcinoma, and provide experimental evidence for its prognosis and biotherapy.Method: The experiments in the thesis adopted in-situ hybridization to measure the expression of let7 and SP immunohistochemistry to measure Ras,HMGA2,C-myc in 68 cases of non-small cell lung carcinoma and 20 cases of normal lung tissue, and analyse their relation to clinic-pathological parameter and survival rate of the non-samll cell lung carcinoma patients.Result: The positive rate of let7,Ras,HMGA2,C-myc in non-small cell lung cancinoma is 39.7%,66.2%,63.2% and 54.4% respectively; and the rate is 75.0% (p<0.05),25.0% (p<0.01),0 (p<0.01),25.0% (p<0.05)in normal lung tissue.(2) The positvie rate of let7 has no relation with patinet's age,gender,histological type,cell differentiation and clinical stages(p>0.05);That of Ras is correlated to smoking and the histological type of carcinoma (p<0.01),and has no relation with cell differentiation,lymphatic metastasis and clinical stages(p>0.05); And the positvie rate of HMGA2 has relation with the tumor size,histological type and lymphatic metastasis(p<0.01),has no relation with age, cell differentiation and clinical stages(p>0.05); The positvie rate of C-myc has no relation with patient's age,gender, histological type, cell differentiation and clinical stages(p>0.05).(3) Spearmen rank presents an obvious negative correlation between let7 and Ras,let7 and HMGA2,let7 and C-myc ; no correlation between Ras and HMGA2,and a positive correlation between Ras and C-myc,HMGA2 and C-myc in non-small cell lung carcinoma.(4).Kaplan-Meier survival analysis shows that the survival rate of positive let7 is higher than that of the negative group; The survival rate of positive HMGA2 is lower than that of the negative group; There has no statistical significance between positive and negative groups of Ras and C-myc..Conclusions: The lower expression level of let7, high expression level of Ras,HMGA2 and C-myc have much to do with the carcinogenesis of NSCLC; The positive expression level of let7 and negative expression level of HMGA2 closely related to prognosis; while the expression of Ras,C-myc may act in cooperativity in the occurrence and development of NSCLC.
