Objective: Model of rats with NASH (non-alcoholic steatohepatitis) were established by feeding with fat-rich diet and injecting oxytetracycline intraperitoneally. To explore the role of IETM (intestinal endotoxemia),TREM-1 and TREM-2 of liver in the pathogenesis of NASH; the effect of glycine on expression of TREM-1 and TREM-2 of rats liver with NASH.Methods: Thirty-six male SD rats after a week of gaving diet freely at about seventeen to twenty-three centi-degree and nature illumination were randomly divided into three groups, twelve every groupâ‘ model group: standardchow+2%cholesterol+151ard+0.5%sodium cholate+5%yolk power, These rats were injected oxytetracycline intraperitoneally about 10mg/100g every five days,the dose of drug is 20mg per 1ml.â‘¡control group:the rats were fed with standard chow and were injected the same volume sodium chloride when model group injected oxytetracycline. (3) treatment group:standardchow+2%cholesterol+151ard+0.5%sodium cholate+5%yolk power+5%glycine.These rats were injected oxytetracycline intraperitoneally about 10mg/100g every five days,the dose of oxytetracycline is 20mg per 1ml. These rats were sacrificed at the 4th,8th weekend during the study. The blood of abdominal aorta was obstained and the levels of plasma TNF-a (tumor necrosis factor-a),ALT,AST,IL-6,IL-10 and the plasma ET of vena portalis hepatis were measured; histopathology of liver tissue was analyzed by light microscope and in the same time the expression of TREM-1,TREM-2 in liver tissue was detected by ELISA.Results:(1)The weight of all rats in three groups increased progressively. The weight and liver index in model group were significantly higher than in control group(P<0.05).(2)The level of ALT and AST in model group have significantly highed compared with the control group at 4th weekend (P<0.05) and significantly depressed in treatment group than in model group(P<0.05).(3)Compared with control group,the plasma TNF-a,ET increased from the 4th week in model group(P<0.05); Compared with model group, they decreased significantly in treatment group at the 8th week(P<0.05).(4) The level of IL-6 in model group were significantly higher than in control group at the 8th week(P<0.05),it decreased significantly in treatment group than in model group; The level of IL-10 in model group significantly decreased from the 4th week (P<0.05). (5)The results of ELISA displayed that the expression of TREM-1 and TREM-2 in model group were significantly different from that in control group (P<0.05); Compared with model group, the expression of TREM-1 in treatment group decreased significantly at the 4th week (P<0.05),while the expression of TREM-2 in treatment group increased from the 4th week and markedly increased at the 8th week(P<0.05).(6) The results of correlation analysis:The result of correlation analysis between liver TREM-lwith plasma TNF-a,ET,IL-6 is significant positive correlation and is negative correlation with IL-10(P<0.01);The result of correlation analysis between liver TREM-2 with plasma ET,TNF-a,IL-6 is significant negative correlation and is positive correlation with IL-10(P<0.01).Conclusion:1,The model of NASH that established by feeding with fat-rich diet and injecting oxytetracycline intraperitoneally was accompanied with IETM. 2,ET played an important role in the genesis and development of NASH by up-regulating expression of TREM-1 and down-regulating TREM-2. 3,Glycine could down-regulate the level of TREM-1 and up-regulate the level of TREM-2 by antagonizing with ET and then lighten the damage of cytokine on liver with NASH, so provided theory proof for the clinical treatment of NASH.
