Objective: To explore the expression and significance of TREM-1、2(triggering receptorexpressed on myeloid cells-1、2)of liver in the(non-alcoholic steatohepatitis, NASH)rats andgallstone patients.Methods: High-fat diet fed rats and intraperitoneally injected a certain amount of oxygentetracycline build NASH rats model.Selected fifty-four clean male SD rats, comfortableenvironment,free diet,divided into three groups after a week,eighteen every group.①controlgroup:normal diet,the volume of saline intraperitoneal injection with the model group wereinjected with oxytetracycline.②model group:ordinary feed mixed with 0.5% sodiumcholate、2% cholesterol、5% yolkpower、15%lard; 10mg/100g the dose every other five daysintraperitoneal injection of oxytetracycline ( each 1ml contains 20mg oxytetracycline ).③intervention group: plus 5% glycine feed on the basis of the model group,the rest of the same tomodel group. Rats were sacrificed at the weekend of the 4, 8, 12, six every group.Measurementof body weight, liver weight, liver index; specimens from liver tissue HE staining to observepathological changes;detection of abdominal aortic plasma TG、TNF-α、PCIII、ALT、AST andportal venous plasma endotoxin (ET)levels;Liver tissue using fluorescence quantitativeRT-PCRmethod to detect mRNA expression levels of TREM-1,TREM-2.Collectting 40 patients that werediagnosed gallstones and no history of excessive alcohol consumption (proposedcholecystectomy) and detectting the plasma aminotransferase (ALT, AST), blood glucose andlipid levels and calculate body mass index, take the size of 0.5×0.5×0.3cm liver in thecholecystectomy (patienthas signed informed consent)to do HE staining,and useimmunohistochemical method to detect the expression of TREM-1, TREM-2 in liver.Results: (1)Model group body weight, liver index with the same period in the control groupwere significantly increased( P <0.05 );(2)Model group rats ALT、AST、TG、PCⅢgroup weresignificantly increased than the control group(P<0.05),the intervention group was lower than thesame period in the model group(P< 0.05);(3)The model group plasma TNF-α、ET increasedcompared with control group(P<0.05); intervention group over the model group was significantlylower (P<0.05);(4)The fluorescent quantitative RT-PCR results showed that the TREM-1mRNAincreased in the model rats compared with control group(P<0.05), TREM-2mRNA decreased inthe model rats compared with control group(P<0.05),the expression of TREM-1mRNAdecreased in the intervention group compared with model group(P<0.05), but TREM-2mRNAincreased(P<0.05);(5)A straight line related to analysis of the results of TREM-1mRNA the expression of the amount and the ET、TNF-α、TG、PCⅢshowed positive related( P<0.01);TREM-2mRNA the expression amount of the ET、TNF-α、TG、PCⅢshowed anegative(P<0.01);(6) Gallstone patients with fatty liver of ALT、AST、TG、blood glucose andbody mass index the proportion of increased people are higher than patients without fatty liver(P<0.05);The difference of cholesterol in gallstone patients with fatty liver the proportion ofincreased people was no significant with patients without fatty liver;(7)Immunohistochemistryshowed that gallstones with fatty liver patients TREM-1 and TREM-2 expression levelscompared with non-fatty liver patients was significantly (P<0.05);(8)TREM-1 protein expressionlevels with the aggravation of the severity of fatty liver and increased, TREM-2 proteinexpression levels with the aggravation of the severity of fatty liver and decreased.Conclusion:1、Accompanied intestinal endotoxemia(IETM )in NASH rat model byhigh-fat diet fed rats were intraperitoneally injected with a certain dose of oxytetracyclineestablished.2、Endotoxin make the expression of TREM-1 increased TREM-2decreased,resulting in TREM-1/TREM-2 ratio imbalance, play an important role in NASH withthe liver damage.Glycine could reduce endotoxin levels make the expression of TREM-1reducedin liver tissue and TREM-2 expression increased, thereby reducing the NASH, disproveTREM-1/TREM-2 and IETM ’s role in NASH.3、The expression of TREM-1 in gallstones withfatty liver patients was increased, TREM-2 was decreased, consistent with these animalexperimental results, provided a theoretical foundation to further investigate the pathogenesisand prevention of NASH.
