Objective:This experiment, by investigating and comparing effects of Perfluorocarbon liquid machine perfusion(PLMP) preservation of donor liver in different temperature conditions, aims to explore the influence of temperature on the process of PLMP preservation of donor liver, so as to unearth the optimal temperature of PLMP preservation of donor liver.Methods:30 male Wistar rats were randomly divided into three groups averagely:one contrast group (Contrast Group) and two experimental groups (Group 1 and Group 2). Contrast Group (n=10):Donor liver was dealt with the machine perfusion. That is the liver was preserved in HC-A mixed with perfluorocarbon liquid for 8 hours in 4 degrees centigrade, at a perfusion speed of 0.2ml·min-1·g-1.Having been preserved, the liver was dealt with Isolace Perfused Rat Liver (IPRL) model, which was perfused by KH. And it was filled with pure oxygen using 100 percent density, at a perfusion speed of 0.2ml·min-1·g-1.The temperature was 37 degrees centigrade and the time for perfusing was 60 minutes. Group 1:Also experimental group 1(n=10), the liver was dealt with the machine perfusion. It was preserved in HC-A mixed with perfluorocarbon liquid for 8 hours in 10 degrees centigrade at the above speed. The liver was also processed with IPRL model after perfusing. Group 2:Also experimental group 2(n=10), the liver was dealt with the machine perfusion. It was preserved in HC-A mixed with perfluorocarbon liquid for 8 hours in 16 degrees centigrade at the same speed. The liver was also processed with IPRL model after perfusing. And Before perfused, each group was added with hyaluronic acid(HA) which had the density of 300ng/ml in the KH. When the livers were perfused for 10 minutes,30 minutes and 60 minutes separately,2 ml's incoming perfusing liquid and 6 ml's effluent liquid were both collected, which were used to test the changes of alanine aminotransferase (ALT), tumor necrosis factor(TNF-a) and hyaluronic acid(HA) absorption in perfusion solution. After the IPRL process, getting some hepatic left medial histology, the morphologic changes of the liver cell and Sinusoidal endothelial cell (SEC) were both observed under light microscopy and electron microscopy.Results:Compared with Contrast Group, Group 1 and 2 had higher ALT activity and TNF-a content, lower HA absorption, and all those differences had statistics significances (P<0.05). Compared Group 2 with Group 1, ALT activity and TNF-a content were both higher in Group 2, HA absorption was lower, which all had significant differences (P<0.05). Furthermore, the histopathologic changes were aggravated, including that the liver cell obviously swollen, vacuolar degeneration, punctiform necrosis, and SEC swell, degeneration, configuration destroyed in the Group 1 and 2, with Group 2 worsening. In addition, SEC was also swollen and degenerated in Contrast Group, but the change in the Contrast Group was obviously lighter than the experimental groups.Conclusion:The effect of PLMP preservation is best when the temperature is 4℃.
