Yu-ping-feng Power, a Chinese traditional prescription, is widely used in China for the prevention and treatment of diseases related to immunologic disorder. However, as the chemical components are complicated, the content determination method has not been established in the original standard. The qualitative or quantitative way to evaluate the quality Chinese traditional medicine mostly which according to only one or few chemical composition reduces the identification accuracy and specificity, and fingerprint analysis method is the control method that provide a reliable guarantee for YPF and its quality ingredients.The fingerprint of Chinese traditional medicine is a new method to analyze the quality Chinese traditional medicine (mainly secondary metabolites) by denoting and describing its chemical information in the image with the modern analytical and detection techniques. It is the control method that reflect the quality of the Chinese traditional medicine preparation and recognize by the international. It provides a comprehensive, macroscopic and quantifiable quality assessment for medicines and preparations using fuzzy and integrity of its basic properties, that is, the uniqueness of species characteristics and the same kind of similarity between individuals. It includes chromatographic fingerprints, spectral fingerprints, fingerprint capillary electrophoresis, NMR fingerprint, fingerprint mass spectrometry, X-ray diffraction fingerprints and DNA fingerprints and so on, in which HPLC chromatographic fingerprint is of the majority. It works as the most important detection method to control quality Chinese traditional medicine with its wide application, excellent separation effect and detection accuracy.This paper includes two parts:Part one: The establishment of the HPLC fingerprint of YPF water extractionA novel, efficient and accurate fingerprint method based on high-performance liquid chromatography-photodiode array detection (HPLC-DAD) for the quality control of the Yupingfeng Power was described and optimized. This part experiment compared the separation effect to YPF water extraction influenced by the different mobile phase system of methanol-water, acetonitrile-water, methanol-0.2% phosphoric acid and acetonitrile-0.2% phosphoric acid and the different detection wave-length, optimized the chromatographic separate conditions by gradient elution, with Prim-O- glucosylcimifygin and 4, -O-β-D-glucosyl-5-O-methylvisamminol and formononetin as index, and build the HPLC fingerprint of YPF. And the 8 peaks that ascertained as the common peaks of the fingerprint form its unique fingerprint. Comparing with the reference peak of Prim-O-glucosylcimifygin, the relative retention time, the relative areas of other peaks and the similarity degree of each fingerprint are calculated as the technique parameters to evaluate the fingerprint. As a result, the similarity degree in 10 batches YPF picking in different time is very different, to some extent, it reflects the content of the active components in different various batches. Part two: The establishment of the HPLC fingerprint of rats, blood serum after the administration YPF water extraction and pharmacokinetics of partial componentsBased on the chromatographic conditions of a stable and efficient HPLC fingerprint of YPF, this part of the experiment which was used deproteinization and liquid-liquid extraction method in serum samples established serum HPLC fingerprints at different times after the administration YPF water extraction in rats. It explains the conversion procedure after medicine into the body and makes the basis of efficacy study of medicine by comparison with fingerprints of medicine, blank serum and serum samples and determination composition and content of the blood at different times.Based on the results, this part researched certain peaks characteristics specifically, finally further determined the blood content, drew the peak concentration/peak areas-time curve and made experimental data for pharmacokinetic study.
