Objective This study investigates the effects of citral on acute promyelocytic leukemia cell lines NB4 cell growth inhibition and induction of apoptosis, and lays the foundation for further study.Method The effects of different concentrations of citral on NB4 cells proliferation was determined by CCK-8 assay. The NB4 cell shape changes after citral impact were observed with optical microscopy. DNA ladder formation was used to detect NB4 cell DNA rupture. FCM was used to measure the proportion of early apoptosis under different citral concentrations.Results Citral inhibits NB4 cell growth. The effect is positively related to the time and dose. After applying 10μg/ml and 20μg /ml citral for 16 hours, optical microscopy observed that the cells show chromatin condensation, marginalization, karyolemma disintegration, chromatin segmentation and apoptotic bodies,and other typical forms of apoptosis. DNA fragment experiments found typical DNA ladder formation. FCM results show that the rate of cell apoptosis increases as citral concentration increased gradually.Conclusion Citral can inhibit the NB4 cell growth and proliferation. Its effect is positively relative to the concentration and duration of time. Citral can also induce NB4 cell apoptosis. The percentage of apoptosis is positively related to the concentration and duration of time.
