ObjectiveThis paper is to construct FAK RNAi lentiviral vector and explore its effect to the activity of gastric cancer cell proliferation, invasion and metastasis ability , which provides a basis for a new gene therapy in gastric cancer.Methods1.To construct FAK shRNA lentiviral vector, its efficiency of gene transfer was detected by the green fluorescent protein reporter gene.2. Gastric cancer SGC7901 cells were infected by FAK shRNA lentiviral vector, MTT was employed to assay the proliferation of SGC7901 cells and Western blot was performed to detect the expression of FAK after infection in SGC7901 cells.3. To prepare Transwell chambers to detect the influence of FAK silencing on SGC7901 cell invasion and metastasis.Results1. To construct successfully targeting FAK RNAi lentiviral vector ,which can infect stably gastric cancer cells .2. The expression of FAK protein level was found to reduce by Western blot in interfere SGC7901 gastric carcinoma cells compared with untreated group and negative plasmid control group. MTT showed that growth of the cells was inhibited in interference group, but not in untreated group and negative control group (P <0.05). 3.Invasion in vitro experiments: To compare the untreated group and negative control group cells, the ability of cell invasion and metastasis decreased significantly in the treated group (P <0.05), the normal group and negative control group had no significant difference (P>0.05)Conclusions1. Constructing successfully FAK RNAi lentiviral vector.2. Reconstructing lentiviral vector effectively inhibited gastric cancer SGC7901 cells transfected with proliferative activity.3.The gastric cancer SGC7901 cell invasion and metastasis were significantly inhibited by FAK silencing.
