Study On The Significances And Expression Of FAK, MadCAM-1 And β7 In Gastric Carcinoma

Objective To explore the influence of expressions of FAK, MadCAM-1 andβ 7 cell on gastric carcinoma during the process of arising and development of gastric carcinoma.Method By immunohistochemical method, the expressions of FAK, MadCAM-1 and β7 were studied in gastric carcinoma, normal gastric tissues and cancer-adjacent tissues of 50 collected specimens when carried out radical resection of gastric carcinoma. And also the expression of FAK in 200 regional lymph nodes was observed.Results The overexpression rates of FAK were 10% (5/50) , 20% (10/50) and 78% (39/50) in normal gastric mucosae, cancer-adjacent tissues and gastric carcinoma tissues respectively. The overexpression of FAK in gastric carcinoma tissues was obviously higher than in normal gastric mucosae and cancer-adjacent tissues (P<0.01). There are no obvious difference between normal gastric mucosae and cancer-adjacent tissues (P>0.05). The strong positive expression rates of FAK were 68.0%( 22/32 )and 33.3%(6/18)in metastasis and no metastasis groups of gastric carcinoma respectively. The overexpression of metastasis group was obviously higher than no metastasis group(JP<0.05). The overexpression rates of FAK were 87.1% (136/156) and 40.9%(18/44) in groups of metastasis lymph node and non-metastasislymph node. The overexpression of metastasis lymph node group was obviously higher than no metastasis group (P<0.01) . The overexpression rate of FAK shows correlation with the differentiation grade and the invasive depth of gastric carcinoma. The deeper invasive and the lower differentiation, the stronger expression rate was obtained(P<0.05). And the overexpression of FAK show no correlation with Borrmarn type, tumor location and tumor size(P >0.05). The expressions of MadCAM-1 were 78%(39/50), 64%(32/50) and 40%(20/50) in normal gastric mucosae, cancer-adjacent tissues and gastric carcinoma tissues respectively. The expression rate of MadCAM-1 in gastric carcinoma tissues was higher than in normal gastric mucosae and cancer-adjacent tissues (P<0.01), and no obvious difference in normal gastric mucosae and cancer-adjacent tissues (P>0.05). The expression rate of MadCAM-1 was lower in young group than old group. The expression of FAK show no correlation with In Borrmarn type,tumor location, tumor size and the differentiation and infiltration grade of gastric carcinoma.(P>0.05). There are different numbers of £ 7 immunological cells between MadCAM-1 positive and negative group. The numbers of (3 7 in MadCAM-1 positive group were obviously more than in negative group. In MadCAM-1 positive control the numbers of P 7 in gastric carcinoma tissues were less than in normal gastric tissues and cancer-adjacent tissues. Conclusion The expression of FAK relate with the process of carcinogensis and development of gastric carcinoma. The increased level of FAK mainly caused by the invasion and metastasis grade of gastric carcinoma cells. Studied FAK expression in cancer tissues can be helpful to understand the carcinogenic biological behavior ofgastric cancer as well as to make right judgment of treatment and prognosis of patients. MadCAM-1 participated in the local immunity of cancer. The low expression of MadCAM-1 may decrease the immune competence and induce carcinogenesis of gastric carcinoma. The expression of MadCAM-1 related with the numbers of P 7 immunological cell. The expression of MadCAM-1 and the change of 3 7 immunological cell may be important molecule incident in the carcinogenesis and development of gastric carcinoma. Perhaps, increase the immunoreactivity of MadCAM-1 is a good immunology method for healing gastric cancer.
