Objective To study the neuroprotective effects of neuregulin-1β(NRG-1β) after cerebral ischemic reperfusion in rats.Methods 54 adult male rats were divided randomly into NRG-1βgroup,NS group and sham group. Nylon Monofilament was applied to establish the middle cerebral artery occlusion reperfusion models(MCAO/R)in the rats. The rats in each group were sacrificed at 1 d, 3 d and 7d after MCAO/R. The nervous behavioral function was evaluated with Zea longa score and inclined plate experiment after the rat wake one hour. RT-PCR was used to detect the expression of GAP-43 mRNA in in the gray matter around the cerebral infarct of rats.Results Comparing with the NS group, NRG-1βgroup can improve the behavior function on the 7 day according to Zea longa score. The electron microscopy showed that NRG-1βgroup were less damaged. The immunohistochemistry showed that NRG-1βgroup expressed more GAP-43 protein. RT-PCR also showed that the expression of GAP-43 mRNA in gray matter of rats were obviously increased in NRG-1βgroup compared with controlled group and sham group(p<0.05 ).Conclusion Neuregulin-1βmay have neuroprotection by improving the nervous behavioral founction and the expression of GAP-43 mRNA in gray matter around the cerebral infarct of rats.
