Objective:Detect the effect of different concentration aspirin(5mmol/L,10 mmol/L,20mmol/L) on the multiplication enterocyte Caco-2 cells,the mucosal barrier of Caco-2 cells,the expression of occluding and observe the ultrastructural organization of the tight junction of Caco-2 cells, and investigate the mechanism of Aspirin on the enterocyte .Method:1. After co-culturing of aspirin solution and Caco-2 cells for 24 hours and 48 hours, the proliferation of Caco-2 cells were examined by MTT.2. In the model of Caco-2 cells, The cells were dealed with aspirin of different concentration, Transep ithelial resistance ( TER) was measured using an EVOM voltohmmeter.3. Use Immunohistochemistry (EnVision method) to detect the expression of occludin of Caco-2 cells dealed with Aspirin of different concentration.4. The injury of Caco-2 cells and the changes in the morphology of TJ stuctures were observed by transmission electron microscopy.Results:1. The restrain effect of aspirin on the proliferation of Caco-2 cells was dose-dependent(5mmol/L,10mmol/L,20mmol/L) and time-dependent.2. The destroy of aspirin on the epithelial barrier of Caco-2 cells was dose-dependent(5mmol/L,10mmol/L,20mmol/L) and time-dependent.3. The expression of Occludin was dose-dependent, and it disappeared in the TJ and emerge in the endochylema when the concentration was 20mmol/L.4. The destroy of Aspirin on the cell organ,cell integrity and the intercellular space(ICS) were dose-dependent.Conclusion:Aspirin can restrain the proliferation of enterocyte and affect the epithelial barrier, and this two effects were dose-dependent and time-dependent, and it can cause the cell-damage directly.
