Objective:To survey the expression of semaphorin 3A(SEMA3A) and neuropilin-1(Nrp-1) in vivo and vitro, so as to the approach the significance of angiogenesis in squamous cell carcinomar of tongue(tongue carcinomar).Method:The expression of SEMA3A and Nrp-1 in human carcinoma of tongue specimens, normal tongue tissue and transplanted tumor in nude mice was investigated by immunohistochemical staining and which was also investigated by immunocytochemistry in cell line Tca8113. The expression of SEMA3A and Nrp-1 was examined in carcinoma of tongue specimens, normal tongue tissue and cell line Tca8113 using Western blotting and RT-PCR. The data was analyzed with SPSS 11.0 software package forχ2 test.Results: Immunohistochemical analysis revealed that Semaphorin 3A was absent in 17 of 20 human carcinoma of tongue specimens while Neuropilin-1 was strongly experssed in 18 of 20. SEMA3A was detected in 19 of 20 normal tongue tissues while Nrp-1 was absent in all of them. And in transplanted tumor immunohistochemical analysis revealed that the expression of semaphorin 3A was absent while Neuropilin-1 was strongly expressed, and the expression has no significant difference at different stages(P>0.05). Immunocytochemistry in cell line Tca8113 suggested the expression of Nrp-1 was positive while SEMA3A was absent. Western blotting and RT-PCR supported the result. The expression of SEMA3A and Nrp-1 in human carcinoma of tongue specimens and normal tongue tissue has significant difference( P<0.01).Conclusion:SEMA3A and Nrp-1 was expressed abnormally in carcinoma of tongue specimens and cell line Tca8113. It suggests that the abnormal expression of SEMA3A and Nrp-1 maybe have relationship with angiogenesis in carcinoma of tongue.
