Hepatocarcinoma is one of the most common fetal diseases all around the world, seriously harm human health. At present, for the majority of hepatocarcinoma, chemotherapy is still the most commonly used treatment. Rapidly development of tumor resistance to chemotherapy drugs represents a major problem to successful cancer treatment. Studies have shown that hepatocarcinoma resistant to apoptosis can be inhibited, and the genes related to apoptosis regulation may lead to abnormal expression of hepatocarcinoma to chemotherapeutic drug resistance. Elucidating the mechanism of this complex process may prevent and reverse the drug resistance, and thus may serve as targets for therapeutic, diagnostic tests or alternatively predictive markers.Our previous study suggested that OPN3, a homologue of insecticide resistance opsin gene in Cx. pipiens pallens, participated in the 5 - fluorouracil resistance of the hepatocarcinoma cell line Bel-7402.In this study, the quantitative PCR was used to detect and verify the cell transfection results. Subsequently, using flow cytometry, we analyzed the cell apoptosis by Annexin V / Propidium Iodide (PI) staining, suggesting that OPN3 may upregulate the cell apoptosis signal pathway.At the same time, to explore the mechanism of apoptosis, Western blot was used to verify flow cytometry experiment and detect the expression of some protein in signal pathway. The results showed that OPN3 could regulate the expression of Akt/p-Akt/Bcl-2/Bax signal pathway.The positive regulation of apoptosis suggested that OPN3 could be the new target for tumor gene therapy. And this work may improve the research of mechanism for hepatocarcinoma resistant to chemotherapy drugs.
